摘 要:目的本研究对4对胃癌患者癌组织及癌旁组织的lncRNA和mRNA进行表达谱定量分析,通过生物信息学方法挖掘到潜在胃癌相关的靶标分子。方法利用基因技术筛选样品得到差异lncRNA和mRNA,对差异lncRNA和mRNA构建共表达网络图,对候选lncRNA利用UCSC重新注释,预测lncRNA结合TF和蛋白,利用GO和KEGG数据库富集分析lncRNA共表达基因和结合蛋白从而推测lncRNA参与功能。结果共筛选出了543个差异lncRNA以及2705个mRNA,GO功能分析显示lncRNA共表达基因参与transferase activity、immune system process、regulation of cell differentiation、immune response、cell differentiation,KEGG通路富集分析显示lncRNA共表达基因参与transferase activity、cell differentiation、p53 signaling pathway、TGF-beta signaling pathway通路。ENST00000444102是一个保守lncRNA,胃癌中显著上调参与肿瘤通路过程。结论ENST00000444102这条lncRNA参与多种肿瘤相关通路,可能成为潜在的胃癌预后、诊断和治疗的治疗靶点。lncRNA参与多种肿瘤相关通路ENST00000444102,可能成为潜在的胃癌预后、诊断和治疗的治疗靶点。Objective In this study,the expression profiles of lncRNA and mRNA in cancer tissues and adjacent tissues of 4 pairs of patients with gastric cancer were quantitatively analyzed,and potential gastric cancer-related target molecules were mined through bioinformatics methods.Methods Samples were screened using gene technology to obtain differential lncRNA and mRNA.Co-expression network maps were constructed for differential lncRNA and mRNA.Candidate lncRNA were reannotated using UCSC to predict lncRNA binding TF and proteins.The co-expressed genes and binding proteins of lncRNA were enriched and analyzed using GO and KEGG databases to speculate the role of lncRNA in function.Results A total of 543 different lncRNA and 2,705 mRNA were screened.GO functional analysis showed that lncRNA co-expressed genes were involved in transferase activity,immune system process,regulation of cell differentiation,and immune response and cell differentiation.KEGG pathway enrichment analysis showed that lncRNA co-expressed genes were involved in transferase activity,cell differentiation,p53 signaling pathway,and TGF-beta signaling pathway.ENST00000444102 is a conserved lncRNA that is significantly upregulated and involved in tumor pathway processes in gastric cancer.Conclusion ENST00000444102 is involved in a variety of tumor-related pathways and may be a potential therapeutic target for the prognosis,diagnosis and treatment of gastric cancer.
