摘 要:目的:探讨阿可拉定(Icaritin)体内体外抗宫颈癌作用及对自噬蛋白p62表达的影响。方法:⑴体外培养宫颈癌Siha、Hela细胞,采用CCK-8实验方法,选用不同浓度的阿可拉定(0、5、10、15、20及50μmol/L),对宫颈癌细胞系Siha、Hela进行24、48、72、96 h阿可拉定药物处理,检测对肿瘤细胞增殖的影响;采用流式细胞、Western Blot法,观察阿可拉定对Siha、Hela细胞凋亡作用以及自噬蛋白p62表达的影响。⑵动物水平探讨阿可拉定抗宫颈癌作用效果:建立裸鼠宫颈癌异种移植瘤模型,研究阿可拉定对异种移植瘤的作用及对p62表达的影响。结果:⑴体外实验发现,阿可拉定可促进宫颈癌细胞Siha、Hela细胞凋亡(P<0.05),同时显著下调p62蛋白的表达(P<0.05)。⑵体内实验中,阿可拉定对人子宫颈癌异种移植瘤的生长具有抑制作用(P<0.001),下调小鼠异种移植瘤中p62表达(P<0.01)。结论:阿可拉定可在体外抑制宫颈癌细胞增殖、促细胞凋亡,体内抑制宫颈癌异种移植瘤生长,可能与显著下调p62蛋白表达有关。Objective To investigate the in vitro and in vivo anti-cervical cancer effects of akoladine and its effect on the expression of autophagy protein p62.Methods⑴Cervical cancer Siha and Hela cells were cultured in vitro,and CCK-8 experimental method was used.Different concentrations of akoladine(0,5,10,15,20,and 50μmol/L)were selected to treat cervical cancer cell lines Siha and Hela for 24,48,72,and 96 hours.Flow cytometry was used to detect the effect on tumor cell proliferation,and Western Blot were used to observe the effect of akoladine on Siha and Hela cell apoptosis and the expression of autophagy protein p62.⑵Exploring the anti-cervical cancer effect of akoladine at the animal level:establish a xenograft tumor model of cervical cancer in nude mice,and study the effect of akoladine on the xenograft tumor and the expression of p62.Results⑴In vitro experiments found that akoladine could promote the apoptosis of cervical cancer cells Siha and Hela(P<0.05),while significantly downregulating the expression of p62 protein(P<0.05).⑵In vivo experiments,akoladine showed inhibitory effects on the growth of human cervical cancer xenograft tumors(P<0.001)and downregulated the expression of p62 in mouse xenograft tumors(P<0.01).Conclusion Akoladine can inhibit the proliferation and promote apoptosis of cervical cancer cells in vitro,and inhibit the growth of cervical cancer xenograft tumors in vivo,which may be related to the significant down-regulation of p62 protein expression.
