摘 要:目的 探讨不同配伍比例的“黄芪-黄连”药对干预2型糖尿病大鼠的作用及对胰岛β、α细胞的影响。方法 从40只SPF级Wistar雄性大鼠中随机选6只作为空白组,其余大鼠采用高糖高脂饲料喂养12周+腹腔注射35 mg/kg链脲佐菌素方法复制2型糖尿病模型。将造模成功的大鼠随机分为模型组、“黄芪-黄连”1∶1组、“黄芪-黄连”3∶1组和拜糖苹组,每组8只。“黄芪-黄连”1∶1组给予黄芪、黄连各30 g水煎液灌胃,“黄芪-黄连”3∶1组给予黄芪45g、黄连15 g水煎液灌胃,拜糖苹组给予拜糖苹60 mg/(kg·d)悬液灌胃,空白组和模型组给予等量蒸馏水灌胃,均干预8周。观察各组大鼠一般状态,检测各组大鼠空腹血糖、糖化血红蛋白、血清胰高血糖素水平,免疫荧光共染观察胰腺组织β细胞、α细胞分布情况,计算β/α细胞比值。结果 灌胃干预后,“黄芪-黄连”3∶1组大鼠一般状况恢复最好并接近于空白组;“黄芪-黄连”3∶1组空腹血糖、糖化血红蛋白、血清胰高血糖素水平和拜糖苹组空腹血糖、糖化血红蛋白水平均明显低于模型组(P均<0.05),“黄芪-黄连”1∶1组仅空腹血糖水平明显低于模型组(P<0.05);免疫荧光染色显示,“黄芪-黄连”1∶1组、“黄芪-黄连”3∶1组及拜糖苹组胰岛内部β细胞数量较模型组增加,且排列规则有序,β/α细胞比值均明显高于模型组(P均<0.05)。结论 “黄芪-黄连”配伍具有良好降糖作用,可有效恢复胰岛β细胞的数量和排列及胰岛内β/α细胞比值,其中以“黄芪-黄连”3∶1配伍比例干预效果佳。Objective It is to investigate the effects of two different combinations of “Astragalus-Coptis” on type 2 diabetes mellitus(T2DM) rats and their effects on β and α cells of pancreatic islets.Methods Six rats were randomly selected from 40 SPF-grade Wistar male rats as a blank group,and the rest rats were fed with high-sugar and high-fat diet for 12 weeks+intraperitoneal injection of 35 mg/kg streptozotocin to replicate T2DM models.The successfully modeled rats were randomly divided into model group,“Astragalus-Coptis” 1∶1 dose group,“Astragalus-Coptis” 3∶1 dose group,and Balsamina group,with 8 rats in each group.The “Astragalus-Coptis” 1∶1 dose group was given water decoction containing Astragalus 30 g and Coptis 30 g by gavage,the “Astragalus-Coptis” 3∶1 dose group was given water decoction containing Astragalus 45 g and Coptis 15 g by gavage,and the Balsamina group was given Balsamina solution 60 mg/(kg·d) by gavage,the blank group and model group were given equal amounts of distilled water by gavage,all the groups were treated for 8 weeks.After 8 weeks of treatment,the general state of rats in each group was observed,the levels of fasting blood glucose,glycosylated hemoglobin,and serum glucagon were detected,and the distribution of β-cells and α-cells in pancreatic tissues was observed by immunofluorescence co-staining,and the β/α-cell ratio was calculated.Results After treatment,the general state of rats in the “Astragalus-Coptis” 3∶1 dose group recovered the best and was close to that of the blank group.The levels of fasting blood glucose,glycated hemoglobin,serum glucagon in the “Astragalus-Coptis” 3∶1 dose group and the levels of fasting blood glucose,glycated hemoglobin in the Balsamina group were all significantly lower than those in the model group(all P0.05),while only the level of fasting blood glucose in the “Astragalus-Coptis” 1∶1 dose group was significantly lower than that of the model group(P0.05),immunofluorescence staining showed that the num
