姜黄素对高糖环境下大鼠肾小管上皮细胞凋亡影响

摘  要:目的 探讨姜黄素对高糖环境下大鼠肾小管上皮细胞凋亡的影响。方法 取对数生长期的大鼠肾小管上皮细胞,实验分为4组:对照组培养基中加入5.5 mmol/L D-葡萄糖;高糖组培养基中加入30 mmol/L D-葡萄糖;姜黄素低剂量组培养基中加入30 mmol/L D-葡萄糖和5μmol/L姜黄素;姜黄素高剂量组培养基中加入30 mmol/L D-葡萄糖和10μmol/L姜黄素。应用流式细胞仪检测细胞凋亡情况,Western blot和实时荧光定量PCR法检测细胞中Bax、Bcl-2蛋白及mRNA表达情况。结果 与对照组比较,高糖组肾小管上皮细胞凋亡率明显增高(P<0.05);肾小管上皮细胞中Bax蛋白及mRNA相对表达量均明显增高(P均<0.05),Bcl-2蛋白及mRNA相对表达量均明显降低(P均<0.05)。与高糖组比较,姜黄素低、高剂量组细胞凋亡率均明显降低(P均<0.05),肾小管上皮细胞中Bax蛋白及mRNA相对表达量均明显降低(P均<0.05),Bcl-2蛋白及mRNA相对表达量均明显增高(P均<0.05),且姜黄素高剂量组各指标改变较姜黄素低剂量组更明显(P均<0.05)。结论 姜黄素可以抑制高糖环境下肾小管上皮细胞凋亡。Objective It is to investigate the renoprotective effects of curcumin on apoptosis of renal tubules epithelial cells under high glucose environment.Methods Rat renal tubular epithelial cells at logarithmic growth period were taken,and divided into 4 groups:control group cultured in the medium added with 5.5 mmol/L D-glucose,high glucose group cultured in the medium added with 30 mmol/L D-glucose,curcumin low-dose group cultured in the medium added with 30 mmol/L D-glucose and 5μmol/L curcumin,and curcumin high-dose group cultured in the medium added with 30 mmol/L D-glucose and 10μmol/L curcumin.the apoptosis of the cells were detected by Flow cytometry,and the expressions of Bax and Bcl-2 protein and mRNA in cells were respectively detected by Western blot and real-time fluorescence quantitative PCR.Results Compared with the control group,the apoptosis rate of renal tubular epithelial cells in the high glucose group was significantly increased(P<0.05),the expressions of Bax protein and mRNA were significantly increased(both P<0.05),the expressions of Bcl-2 protein and mRNA were significantly decreased(both P<0.05).Compared with the high glucose group,the apoptosis rates were significantly reduced in both curcumin low and high dose groups(all P<0.05),the relative expressions of Bax protein and mRNA in renal tubular epithelial cells were significantly reduced(all P<0.05),and the relative expressions of Bcl-2 protein and mRNA were significantly increased(all P<0.05),and the changes of each index in the curcumin high dose group were more significant than that in the curcumin low dose group(all P<0.05).Conclusion Curcumin can inhibit renal tubular epithelial cell apoptosis in a high glucose environment.
