摘　　要：目的:检测TRIM59在结直肠癌及癌旁正常组织中的表达,分析其表达变化与结直肠癌临床病理特征及预后的相关性,探讨其在结直肠癌中的作用。方法:采用实时荧光定量PCR和免疫印迹法检测8份结直肠癌组织及其癌旁组织中TRIM59 mRNA和蛋白的表达情况。通过免疫组织化学法检测30例结直肠癌组织及16例癌旁组织中TRIM59蛋白的表达情况,比较两种组织中TRIM59蛋白的表达差异。收集NCBI的基因表达汇编(GEO)中的编码基因表达谱数据集,对结直肠癌肿瘤样本表达谱资料及其对应的生物学信息进行相关性和预后分析。结果:结直肠癌组织中TRIM59 mRNA相对表达量为0.0956±0.0111,高于癌旁组织的0.0239±0.0162,差异有统计学意义(t=9.31,P=0.011)。免疫印迹结果表明TRIM59在癌组织中的表达高于癌旁组织。免疫组化结果表明,TRIM59在结直肠癌组织的阳性表达率为86.7%(26/30),高于癌旁正常组织的25%(4/16),差异有统计学意义(U=67,P<0.001)。在结直肠癌中,高表达TRIM59的样本与Dukes分期相关(H=13.067,P=0.004);TRIM59高表达的结直肠癌患者预后更差(P<0.05)。结论:TRIM59在结直肠癌组织中的表达较癌旁组织高,且与结直肠癌的分期和生存率均有关,TRIM59可能参与了结直肠癌的进展。Objective:To detect the expression of tripartite motif containing protein 59(TRIM59)in colorectal carcinoma tissues and corresponding normal colorectal tissues,to explore the relationship between the expression of TRIM59 and the pathologic features and prognosis,and to discuss the role of TRIM59 playing in the colorectal carcinoma.Methods:The expression of TRIM59 mRNA and protein in 8 cases of colon cancer and tumor adjacent tissues were detected by real‑time polymerase chain reaction(RT‑PCR)and Western blot.The expression of TRIM59 at protein level in 30 cases of colorectal cancer and 16 cases of tumor adjacent tissues were determined by SP immunohistochemistry(IHC),and the expression difference between the two tissues was compared.GEO datasets were col‑lected and colorectal cancer samples expression profile and clinical information were downloaded.The correlation between gene expression and clinicopathologic features was analyzed by Chi‑square test.Overall survival and disease‑free survival was analyzed by Kaplan‑Meier curve.Results:The relative expression of TRIM59 mRNA in the colorectal cancer tissues was higher than that in tumor adjacent tissues(0.0956±0.0111 vs 0.0239±0.0162;t=13.29,P<0.01).The Western blot results showed that the relative expression of TRIM59 in the colorectal cancer tissues was higher than that in tumor adjacent tissues.IHC showed that the positive expression rates of TRIM59 in colorectal cancer tissues and nonneoplastic epithelia tissues were 86.7%(26/30)and 25%(4/16),respectively,and the difference was statistically significant(U=67,P<0.001).Higher expression of TRIM59 was as‑sociated with Dukes'stage(H=13.067,P=0.004).Higher expression of TRIM59 indicated poor prognosis in colorectal cancer(P<0.05).Conclusion:The expression of TRIM59 in colorectal can‑cer tissues is higher than that in tumor‑adjacent tissues.Furthermore,it is correlated with stage and survival rate.TRIM59 may be involved in the development of colorectal cancer.
