摘  要:目的:探讨阿司匹林(aspirin,又称acetyl salicylic acid,ASA)对人肾癌细胞系ACHN和786-O细胞增殖、迁移、凋亡和自噬的影响。方法:利用CCK-8法检测不同浓度的ASA及ASA联合氯喹(chloroquine,CQ)对ACHN和786-O细胞存活率的影响;应用平板克隆实验检测不同浓度ASA对肾癌细胞克隆形成能力的影响;采用Transwell实验和划痕实验检测肾癌细胞迁移能力;通过Annexin V-FITC/PI实验检测肾癌细胞的凋亡情况;Western blot法检测ASA对肾癌细胞凋亡相关蛋白Bax、Bcl-2及自噬相关蛋白LC3B、Beclin-1和p62表达水平的影响。结果:ASA可显著抑制ACHN和786-O细胞的存活率,随ASA浓度/时间增加细胞存活率显著下降(P<0.05);与对照组相比,ASA显著抑制肾癌细胞的增殖和迁移(P<0.05);ASA处理增加了肾癌细胞凋亡数目,显著抑制了肾癌细胞凋亡相关蛋白Bcl-2的表达,上调Bax表达,并促进自噬相关蛋白LC3B和Beclin-1的表达,抑制p62的表达(P<0.05)。与CQ处理组相比,ASA联合CQ组显著促进了LC3B的表达抑制p62的表达(P<0.05),并显著抑制了肾癌细胞的增殖和迁移并促进其凋亡(P<0.05)。结论:ASA可抑制肾癌细胞的增殖和迁移并促进凋亡和自噬。Objective:To investigate the effects of aspirin(acetyl salicylic acid,ASA)on the proliferation,migration,apoptosis and autophagy of human renal carcinoma cell lines ACHN and 786-O.Methods:CCK-8 method was used to determine the effect of different concentrations of ASA and chloroquine(CQ)combined with ASA on the survival rate of ACHN and 786-O cells,and plate cloning experiment was used to elucidate the effect of different concentrations of ASA on the clonogenesis ability of renal cancer cells.The migration ability of renal cancer cells was detected by Transwell assay and scartch assay,apoptosis of renal cancer cells was detected by Annexin V-FITC/PI assay.The effects of ASA on apoptosis related proteins Bax,Bcl-2 and autophagy related proteins LC3B,Beclin-1 and p62 of renal cancer cells were detected by Western blot.Results:ASA could observably inhibit the survival rate of ACHN and 786-O cells,and the survival rate of ACHN and 786-O cells decreased remarkably with the increase of ASA concentration/time(P<0.05).Compared with the control group,ASA significantly inhibited the proliferation and migration of renal carcinoma cells(P<0.05).ASA treatment increased the number of apoptosis of renal cancer cells,significantly inhibited the expression of apoptosis-related protein Bcl-2,up-regulated the expression of Bax,promoted the expressions of autophagy related proteins LC3B and Beclin-1,and inhibited the expression of p62(P<0.05).Compared with CQ treatment group,ASA combined with CQ treatment group significantly promoted the expression of LC3B and inhibited the expression of p62(P<0.05),and significantly inhibited the proliferation and migration of renal cancer cells and promoted apoptosis(P<0.05).Conclusion:ASA can inhibit the proliferation and migration of renal carcinoma cells and promote apoptosis and autophagy.
