摘 要:目的:探究槐定碱对前列腺癌PC3细胞凋亡的影响,及其促进细胞凋亡可能的作用机制。方法:采用MTT法分析槐定碱对前列腺癌PC3细胞增殖的抑制作用;台盼蓝染色实验检测槐定碱对前列腺癌PC3细胞生长增殖的影响;Hoechst 33342染色法和流式细胞术检测槐定碱对前列腺癌PC3细胞凋亡的影响;Western blot检测槐定碱对前列腺癌PC3细胞凋亡相关蛋白Caspase-3、Bax、Bcl-2及凋亡信号通路蛋白p-JNK、p-p38、p-ERK表达的影响。结果:随着槐定碱浓度的升高,PC3细胞抑制率逐渐升高,作用呈剂量依赖性,细胞增殖受到明显抑制作用(P<0.05);槐定碱组的细胞凋亡率显著升高,Caspase-3、Bax促凋亡蛋白表达显著增高,而Bcl-2抑凋亡蛋白表达则显著降低(P<0.05);MAPK信号通路蛋白p-JNK表达显著升高(P<0.05),p-p38、p-ERK表达无显著性差异(P>0.05);JNK抑制剂SP600125逆转槐定碱对前列腺癌PC3细胞凋亡的促进作用(P<0.05);槐定碱能下调裸鼠瘤体体积、瘤体质量(P<0.05)。结论:槐定碱能抑制前列腺癌PC3细胞增殖,促进其凋亡,具有良好的抗肿瘤活性;其可能通过JNK/MAPK信号通路介导前列腺癌PC3细胞的凋亡。Objective:To investigate the effect of sophoridine on apoptosis of prostate cancer PC3 cells and the possible mechanism of promoting apoptosis.Methods:The inhibitory effect of sophoridine on the proliferation of PC3 cells was analyzed by MTT method.Trypan blue staining assay was used to detect the effect of sophoridine on the growth and proliferation of PC3 cells.Hoechst 33342 staining and flow cytometry were used to detect the effect of sophoridine on apoptosis of PC3 cells.Western blot detected the effect of sophoridine on the expression of Caspase-3,Bax,Bcl-2,and apoptosis signaling pathway proteins p-JNK,p-p38,p-ERK in PC3 cells.Results:With the increase of sophoridine concentration,the inhibition rate of PC3 cells was gradually increased in a dose-dependent manner,and the proliferation of PC3 cells was significantly inhibited(P<0.05).The apoptosis rate of sophoridine group was significantly increased,and the expressions of Caspase-3 and Bax apoptosis-promoting proteins were significantly increased,while the expressions of Bcl-2 anti-apoptosis-inhibiting proteins were significantly decreased(P<0.05).The expression of MAPK signaling pathway protein p-JNK was significantly increased,while the expression of p-p38 and p-ERK had no significant differences(P>0.05).JNK inhibitor SP600125 reversed the promoting effect of sophoridine on apoptosis of PC3 cells(P<0.05).Sophoridine could reduce tumor volume and mass in nude mice(P<0.05).Conclusion:Sophoridine can inhibit the proliferation and promote the apoptosis of PC3 cells in prostate cancer,and has good antitumor activity.It may mediate apoptosis of prostate cancer PC3 cells through JNK/MAPK signaling pathway.
