摘  要:目的:探讨人参皂苷Rd对结肠癌SW480细胞增殖和凋亡的影响及分子机制。方法:实时荧光定量PCR(RT-qPCR)检测结肠癌组织及细胞中lncRNA MAGI1-IT1表达水平;结肠癌细胞SW480分为对照组、低、中、高剂量人参皂苷Rd组、si-MAGI1-IT1组、si-NC组、高剂量人参皂苷Rd+pcDNA-MAGI1-IT1组、高剂量人参皂苷Rd+pcDNA组。四甲基偶氮唑盐比色法(MTT)、流式细胞术评价细胞增殖和凋亡;蛋白质印迹法检测增殖(CyclinD1、p21)、凋亡(Caspase3、Bcl-2、cleaved-Caspase3、Bax)相关蛋白表达。结果:MAGI1-IT1在结肠癌组织中表达水平升高(P<0.05);中、高剂量人参皂苷Rd处理SW480细胞后,增加G_(0)/G_(1)期比例、凋亡率,降低S期比例,降低CyclinD1、Caspase3、Bcl-2蛋白表达水平,增加p21、cleaved-Caspase3、Bax表达水平,MAGI1-IT1表达水平降低,且呈剂量依赖性(P<0.05)。下调lncRNA MAGI1-IT1后,SW480的G_(0)/G_(1)期比例、凋亡率增加,S期比例下降,降低CyclinD1、Caspase3、Bcl-2蛋白表达水平,增加p21、cleaved-Caspase3、Bax表达水平(P<0.05)。上调lncRNA MAGI1-IT1逆转了人参皂苷Rd对结肠癌SW480细胞增殖和凋亡的影响。结论:人参皂苷Rd通过下调lncRNA MAGI1-IT1表达抑制结肠癌SW480细胞的增殖,促进其凋亡。Objective:To investigate the effect of ginsenoside Rd on the proliferation and apoptosis of colon cancer SW480 cells and its molecular mechanism.Methods:Real-time fluorescent quantitative PCR(RT-qPCR)was used to detect the expression of lncRNA MAGI1-IT1 in colon cancer tissues and cells.Colon cancer cells SW480 were divided into control group,low,medium and high dose ginsenoside Rd group,si-MAGI1-IT1 group,si-NC group,high-dose ginsenoside Rd+pcDNA-MAGI1-IT1 group,high-dose ginsenoside Rd+pcDNA group.Tetramethylazolium salt colorimetric method(MTT),flow cytometry to detect cell proliferation activity and apoptosis.Western blot method to detect proliferation(CyclinD1,p21)and apoptosis-related protein(Caspase3,Bcl-2,cleaved Caspase3,Bax)expression.Results:The expression of MAGI1-IT1 in colon cancer tissue was increased(P<0.05).After medium and high doses of ginsenoside Rd were treated with SW480 cells,the ratio of G_(0)/G_(1) phase,apoptosis rate were increased,ratio of S phase was decreased,decreased the protein expression levels of CyclinD1,Caspase3 and Bcl-2,and the expression levels of p21,increased the expression levels of cleaved-Caspase3 and Bax,and the expression of MAGI1-IT1 was decreased,in a dose-dependent manner(P<0.05).After down-regulating lncRNA MAGI1-IT1,the ratio of G_(0)/G_(1) phase and apoptosis rate were increased,ratio of S phase was decreased,decreased the protein expression levels of CyclinD1,Caspase3 and Bcl-2,and increased the expression levels of p21,cleaved-Caspase3 and Bax(P<0.05).Up-regulation of lncRNA MAGI1-IT1 reversed the effect of ginsenoside Rd on the proliferation and apoptosis of colon cancer SW480 cells.Conclusion:Ginsenoside Rd inhibits the proliferation of colon cancer SW480 cells and promotes their apoptosis by down-regulating the expression of lncRNA MAGI1-IT1.
