摘 要:采用低共熔溶剂(Deep eutectic solvent,DES)提取玉米花粉黄酮类化合物,通过单因素实验探究低共熔溶剂种类、摩尔比、含水量、固液比、提取时间、提取温度对黄酮类化合物提取得率的影响。在此基础上利用响应面优化其提取工艺,并对其抗氧化活性、DES回收和再利用性进行分析。结果表明:DES提取工艺最佳条件为四甲基氯化铵与乙二醇摩尔比为1:3,其含水量28%,固液比1:39 g/mL,提取温度49℃,提取时间32 min,黄酮类化合物提取得率为8.912mg/g,是乙醇提取得率的1.57倍。DES提取物的抗氧化能力比乙醇提取物更强。采用石墨烯回收DES,重复使用4次后,提取率约为原始DES的89%。因此,该绿色环保的低共熔溶剂能高效提取玉米花粉黄酮类化合物,且能够保持良好的抗氧化活性。An extremely economical deep eutectic solvent(DES)extraction of flavonoids from maize pollen was developed.The effects of solvent type,molar ratio of solvents,water content,solide-to-liquid ratio,extraction temperature and extraction time on the yield of flavonoids were investigated through single-factor tests.Response surface methodology was used to optimize the extraction conditions.The antioxidant activity of the extracts and the recyclability and reusability of the DES were investigated.the optimum conditions of DES extraction process were as follows:a tetramethyl ammonium chloride-ethylene glycol molar ratio of 1:3,water content of 28%,solid-liquid ratio of 1:39 g/mL,extraction temperature of 49℃,and extraction time of 32 min.The yield of extracted flavonoids reached 8.912 mg/g,which is about 1.57 times that achieved using ethanol extraction.Furthermore,the antioxidant ability of the DES extract was more potent than that of the extract obtained using ethanol. Used DES can be efficiently recovered using graphene (the recovery yield of DES was around 89% of that of the original DES after four reuses). The green deep eutectic solvents can effectively extract flavonoids from maize pollen and maintain good antioxidant activity.
