摘 要:目的:探讨基于长链非编码RNA(long non-coding RNA,lncRNA)分子对结直肠癌患者预后评价的可行性。方法:通过对TCGA数据库中结直肠癌患者转录组数据的系统挖掘,首先根据差异表达分析,筛选出在癌组织中异常表达的lncRNA分子,然后通过单变量Cox回归分析找出与预后显著相关的lncRNA分子,利用这些分子的回归系数构建结直肠癌预后评价系统。结果:大量的lncRNA分子在结直肠癌组织中呈现异常表达,与正常组织相比,分别有793个和495个lncRNA分子在癌组织中升高和降低,单变量Cox回归分析结果显示有6个lncRNA分子与结直肠癌患者的预后存在显著关联(P<0.05)。利用穷举法,结果显示有NUP50-AS1和RP5-1159O4.12个lncRNA分子的单变量回归系数构建的预后分数对于生存期预测性能最佳(AUC=0.71);根据预后评分,高分组和低分组在生存时间比较,差异有统计学意义(P<0.01)。结论:本研究发现基于NUP50-AS1和RP5-1159O4.12个lncRNA分子构成的预后评分能够在一定程度上作为结直肠癌患者预后的生物标志物之一。Objective To explore the feasibility of using long non-coding RNA(lncRNA)molecules for the prognosis evaluation of colorectal cancer patients.Methods We performed a systematic analysis of the transcriptome data of colorectal cancer patients in The Cancer Genome Atlas(TCGA)database.Firstly,we screened out the lncRNA molecules with abnormal expression in the cancer tissue based on differential expression analysis.Then,we identified the lncRNA molecules significantly associated with prognosis through univariate Cox regression analysis.Finally,we constructed a prognostic evaluation system for colorectal cancer based on the regression coefficients of these lncRNA molecules.Results A large number of lncRNA molecules showed abnormal expression in colorectal cancer tissue,with 793 and 495 lncRNA molecules increased and decreased in cancer tissue compared to normal tissue,respectively.Univariate Cox regression analysis identified six lncRNA molecules significantly associated with the prognosis of colorectal cancer patients(P<0.05).Using the exhaustive method,we found that the prognostic score based on the regression coefficients of NUP50-AS1 and RP5-1159O4.1 lncRNA molecules had the best predictive performance for survival(AUC=0.71).According to the prognosis score,there was significant difference in survival time between high group and low group(P<0.01).Conclusions This study found that the prognostic score based on NUP50-AS1 and RP5-1159O4.12 lncRNA molecules can be used as one of the prognostic biomarkers of colorectal cancer patients to a certain extent.
