摘  要:目的 通过检测视神经脊髓炎谱系障碍(NMOSD)患者及非炎症性神经系统疾病(NND)患者脑脊液中核苷酸结合寡聚化结构域样受体蛋白3(NLRP3)、白介素1β(IL-1β)水平,探索NMOSD组患者脑脊液中NLRP3、IL-1β水平与疾病严重程度的相关性。方法 收集2021年10月至2022年10月于郑州大学第一附属医院神经内科住院或课题生物样本库的35例NMOSD首发患者和20例NND患者,将其分为NMOSD组和对照组。分析两组患者临床资料,包括年龄、性别、临床症状、影像学资料和脑脊液检测指标等。并采用扩展残疾状态量表(EDSS)评分对NMOSD组患者入院时疾病严重程度进行评估。采取酶联免疫吸附试验检测脑脊液中NLRP3、IL-1β水平,并分析其水平与EDSS评分之间的相关性。结果 与对照组相比,NMOSD患者脑脊液NLRP3、IL-1β水平显著升高。此外,脑脊液中NLRP3、IL-1β水平与EDSS得分呈正相关。结论 NLRP3、IL-1β可作为NMOSD患者严重程度的生物标志物和治疗NMOSD的潜在靶点。Objective To explore the correlation between the levels of nucleotide-binding oligomerization domain-like receptor protion 3(NLRP3)and interleukin 1β(IL-1β)in cerebrospinal fluid of patients with neuromyelitis optica spectrum disorder(NMOSD)and the severity of the disease by measuring the levels of NLRP3 and IL-1βin the cerebrospinal fluid of patients with NMOSD and patients with non-inflammatory nervous system disease(NND).Methods Thirty-five patients with NMOSD and 20 patients with NND who were hospitalized in the Department of Neurology of the the First Affiliated Hospital of Zhengzhou University from October 2021 to October 2022 were collected and divided into NMOSD group and control group.The clinical data of the two groups were analyzed,including age,gender,clinical symptoms,imaging data and cerebrospinal fluid detection indexes.Expanded disability status scale(EDSS)score was used to evaluate the disease severity of patients in NMOSD group at admission.The levels of NLRP3 and IL-1βin cerebrospinal fluid were detected by enzyme-linked immunosorbent assay,and the correlation between their levels and EDSS score was analyzed.Results Compared with the control group,the levels of NLRP3 and IL-1βin cerebrospinal fluid of patients with NMOSD were higher.In addition,the levels of NLRP3 and IL-1βin cerebrospinal fluid were positively correlated with the score of EDSS.Conclusion NLRP3 and IL-1βcan be used as biomarkers of the severity of optic neuromyelitis and potential targets for the treatment of NMOSD.
