温度对小鼠海马神经元细胞存活素基因表达及细胞活力影响的初步研究

摘  要:目的观察不同温度变化,尤其是高温对神经元细胞中存活素(SVV)基因的表达水平及细胞活力的影响。方法将培养好的小鼠海马神经元细胞(HT22)随机分为4组:对照组在37℃下培养,将3个实验组分别在33℃、39℃及41℃下培养,各组的培养时间分别为3 h、6 h、12 h、24 h。采用CCK-8法检测细胞活力;通过Western blot检测SVV基因的蛋白表达水平;Real-time PCR检测SVV基因的mRNA表达水平。结果与对照组相比,39℃、41℃组细胞活力下降,SVV基因蛋白表达水平及mRNA表达水平升高,差异有统计学意义(P<0.05)。结论高温可对小鼠海马神经元细胞SVV基因的表达水平及细胞活力产生影响。Objective To observe the effect of different temperature,especially high temperature,on the expression of survivin gene and the cell viability in the mouse hippocampal neuron cells.Methods The hippocampal neuron cells(HT22)were divided into four groups:the control group was cultured at 37℃,and the other three experimental groups were cultured at 33℃,39℃and 41℃for 3 h,6 h,12 h and 24 h respectively.The cell viability was observed by CCK-8 method.The protein expression of survivin was detected by Western blot.Real-time PCR was uesd to detect the expression of survivin mRNA.Results Compared with the control group,the cell viability and the expression levels of survivin mRNA in 39℃and 41℃groups decreased.The difference was statistically significant.Conclusion Hyperthermia can effect the expression level of survivin and the cell viability of HT22.
