摘 要:为了筛选野猪体内潜在的乳酸菌(lactic acid bacteria,LAB)并对其生物学特性进行分析,试验以东北地区野猪粪便为样品,利用MRS培养基分离纯化,结合16S rDNA基因鉴定筛选LAB,并对分离株的耐酸和耐胆盐性能进行测定,进一步分析其黏附、凝集和抑菌特性及在动物体内的安全性。结果表明:从野猪粪便中共分离出106株LAB,经过耐酸和耐胆盐复筛,4株菌株(粪肠球菌2株、唾液乳杆菌1株和乳酸片球菌1株)对低pH值和高浓度胆盐具备良好的耐受能力。4株LAB对Caco-2细胞的黏附率在45.16%~66.66%之间,且自凝集率较高(在70%以上),同时对3株致病菌(大肠杆菌K88、猪霍乱沙门菌C500和金黄色葡萄球菌ATCC 25923)具备较强的共凝集能力;4株LAB对3株致病菌均具有较强的抑菌活性;而将LAB连续饲喂小鼠后无不良反应。说明从野猪粪便中筛选到的4株LAB有潜力作为优良微生态制剂的候选菌株。In order to screen for potential lactic acid bacteria(LAB)in wild boars and analyze their biological properties,in the experiment,wild boar feces in Northeast China were used as samples,and MRS medium was used for isolation and purification;combined with 16S rDNA gene identification,LAB was screened.The acid resistance and bile salt resistance of the isolates were determined.Their adhesion,agglutination and bacteriostatic properties and safety in animals were further analyzed.The results showed that a total of 106 isolates of LAB were isolated from wild boar feces.After acid and bile salt resistance rescreening,four isolates(two isolates of Enterococcus faecalis,one strain of Ligilactobacillus salivarius and one strain of Pediococcus acidilactici)had good tolerance to low pH value and high concentration of bile salts.The adhesion rates of the four isolates of LAB to Caco-2 cells ranged from 45.16%-66.66%.Moreover,the self-agglutination rates were high,above 70%,and these isolates had strong coagglutination ability for three pathogenic bacteria.The four isolates of LAB had strong inhibitory activity against Escherichia coli K88,Salmonela enterica serovar Choleraesuis C500 and Staphylococcus aureus ATCC 25923.There were no adverse reactions after continuous feeding of LAB to mice.The results suggested that the four isolates of LAB screened from wild boar feces had the potential to be candidates for excellent microecological preparations.
