摘 要:目的从泰山白首乌的根际土壤中分离出优质真菌,以期得到具有较好药理作用的天然产物。方法将真菌菌株Leptosphaeria sp.CXY48在大米培养基中静置发酵,以活性为导向,利用正、反相硅胶及高效液相色谱等方法进行单体化合物的分离纯化,并利用质谱、核磁共振波谱(nuclear magnetic resonance,NMR)等光谱技术对化合物结构进行鉴定。结果从泰山白首乌根际真菌Leptosphaeria sp.CXY48的次级代谢产物中分离得到4种苯并吡喃酮类化合物,通过光谱和理化性能的分析,确定其为Fusaraisochromanone 2(1),Clearanol I(2),Banksialactone A(3),(R)⁃3,4⁃dihydro⁃4,6,8⁃trihydroxy⁃4,5⁃dimethyl⁃3⁃methyleneisochromen⁃1⁃one(4),对化合物1~4进行了抗菌活性测试,发现化合物1和4对大肠杆菌标准菌株(E.coli ATCC25922)的最小抑菌浓度(minimum inhibitory concentration,MIC)值分别为3.89和70μg/mL,对金黄色葡萄球菌标准菌株(S.aureus ATCC33591)的MIC值分别为187和250μg/mL。结论通过分离Leptosphaeria sp.CXY48的次级代谢产物,获得的4个化合物,其中有2个化合物有一定的抑菌活性。Objective:To isolate High-quality fungi from the rhizosphere soil of Cynanchum bungei Decne in order to obtain natural products with good pharmacological effects,which laid a material foundation for the development of new drugs with high efficiency and low toxicity.Methods:The fungal strain Leptosphaeria sp.CXY48 was fermented in rice medium by static fermentation,and the compounds were isolated and purified using normal and reversed-phase silica gel and high performance liquid chromatography,and the structures of the compounds were identified using mass spectrometry and NMR spectroscopic techniques.Results:Four benzopyrones were isolated from the secondary metabolites of Leptosphaeria sp.CXY48 from Cynanchum bungei Decne,and were identified by spectroscopic and physicochemical property analysis as Fusaraisochromanone 2(1),Clearanol I(2),Banksialactone A(3),and(R)-3,4-dihydro-4,6,8-trihydroxy-4,5-dimethyl-3-methyleneisochromen-1-one(4).The antibacterial activity of compounds 1~4 was tested.It was found that the MIC values of compounds 1 and 4 were 3.89 and 70μg/mLforE.Coli ATCC25922,and 187 and 250μg/mLforS.aureus ATCC33591,respectively.Conclusion:Four compounds are obtained by isolating the secondary metabolites of Leptosphaeria sp.CXY48,two of which show some antibacterial activity.
