全基因组关联分析鉴定绿壳色素含量拷贝数变异

摘  要:为鉴定影响蛋壳色素含量的基因组拷贝数变异,试验以长顺绿壳蛋鸡为研究对象,依据蛋壳表型设置2个试验组:B2 vs B1和B3 vs B1,通过CGH芯片全基因组检测筛选和Ac⁃cuCopyTM多重基因拷贝数检测验证,在全基因组范围内进行鉴定,选择其中12个注释功能基因开展关联验证。结果显示:两个试验组分别检测到绿壳表型差异关联区域22个和24个;腺苷酸环化酶10(ADCY10)基因拷贝数变异与原卟啉色素含量呈极显著负相关(R=-0.424,P=0.001),与胆绿素色素含量呈显著正相关(R=0.299,P=0.018)。研究提示,ADCY10基因可能是影响蛋壳中不同色素含量的关键基因之一。In order to identify genomic copy number variations(CNVs)affecting eggshell pigment content,Changshun blue eggshell chicken was a research object,and two test groups,B2 vs B1 and B3 vs B1,were set according to the eggshell phenotype.The CNVs were screened by CGH microarray detection and verified by AccuCopyTM multiple gene copy number detectionin in genome-wide.Twelve annotated functional genes in the identification area were selected for association verification.The results showed that 22 and 24 differentially associated regions of green shell phenotype were detected in the two groups,respectively.It was found that the copy number variation of adenylyl cyclase 10(ADCY10)gene was significantly negatively correlated with protoporphyrin pigment content(R=-0.424,P=0.001),and positively correlated with biliverdin pigment content(R=0.299,P=0.018).It′s suggested that ADCY10 gene might be one of the key genes affecting the different pigment contents in eggshell.
