摘　　要：目的:探讨lncRNA OIP5-AS1对膀胱癌细胞增殖、迁移及侵袭能力的影响。方法:实验选取人正常膀胱细胞系(SV-HUC-1)和膀胱癌细胞系(EJ、T24、BIU87)为研究对象,采用Real-time PCR检测细胞lncRNA OIP5-AS1的表达。选取lncRNA OIP5-AS1表达较高的T24细胞进行后续实验,设置组别为control组、siRNA-NC组、siRNAOIP5-AS1组、Vector组、OIP5-AS1组,通过CCK-8检测细胞增殖能力,划痕实验检测细胞迁移能力,Transwell实验检测细胞侵袭能力,Real-time PCR检测细胞lncRNA OIP5-AS1的表达,Western blot检测细胞Vimentin、E-Cadherin蛋白表达。结果:与SV-HUC-1细胞相比,lncRNA OIP5-AS1在膀胱癌细胞中的表达明显升高,T24细胞表达量较高(P<0.05)。与control组相比,siRNA-OIP5-AS1组细胞增殖、迁移、侵袭能力明显降低,Vimentin蛋白表达明显降低,E-Cadherin蛋白表达明显升高(P<0.05)。结论:降低lncRNA OIP5-AS1表达可能通过调节上皮间质转化抑制膀胱癌细胞的增殖、迁移和侵袭能力。Objective:To investigate the effect of lncRNA OIP5-AS1 on proliferation,migration and invasion of bladder cancer cells.Methods:Human normal bladder cell line(SV-HUC-1)and bladder cancer cell lines(EJ,T24,BIU87)were selected for the experiment,and the expression of lncRNA OIP5-AS1 was detected by real-time PCR.T24 cells with higher lncRNA OIP5-AS1 expression were selected for subsequent experiments,and were assigned to the control group,the siRNA-NC group,the siRNA-OIP5-AS1group,the Vector group and the OIP5-AS1 group.Cell proliferation was detected by CCK-8;cell migration was detected by scratch assay;Transwell assay was performed to detect cell invasion;real-time PCR was performed to detect the expression of lncRNA OIP5-AS1 in the cells;Western blot was performed to detect the expressions of Vimentin and E-Cadherin proteins in the cells.Results:Compared with SV-HUC-1 cells,the expression of lncRNA OIP5-AS1 was significantly higher in bladder cancer cells,especially higher in T24 cells(P<0.05).Compared with the control group,cell proliferation,migration,invasion and Vimentin expression were significantly lower in the siRNA-OIP5-AS1 group,and the expression of E-Cadherin was significantly higher in the siRNA-OIP5-AS1 group(P<0.05).Conclusion:Lower the expression of lncRNA OIP5-AS1 can significantly inhibit the proliferation,migration,and invasion of bladder cancer cells by regulating epithial-mesenchymal transition.
