摘  要:目的:探讨硫氧还蛋白-1(Srxn1)在血管性认知功能障碍(VCI)大鼠模型发病过程中的变化。方法:将52只健康雄性SD大鼠随机分为5组,对照组6只不予任何处理;假手术组6只仅分离双侧颈总动脉;13只为常规手术组,运用改良2-V0法结扎双侧颈总动脉;Srxn1-siRNA手术组(13只)和核转录因子红系2相关因子2(Nrf2)-siRNA手术组(14只)均采用改良2-V0法结扎双侧颈总动脉,术前24 h于侧脑室分别注射Srxn1、Nrf2基因干扰片段,建立VCI大鼠模型;根据Morris水迷宫定位航行实验评价各组大鼠空间学习记忆能力;检测各组脑脊液中超氧化物歧化酶(SOD)活力、脑脊液丙二醛(MDA)含量、Srxn1浓度,海马组织中Srxn1、Nrf2蛋白表达水平。结果:常规手术组大鼠的逃避潜伏期长于对照组,单位时间内穿越平台的次数、平台象限滞留的时间较对照组大鼠减少,差异有统计学意义(P<0.05);第4、5天,Srxn1-siRNA手术组及Nrf2-siRNA手术组大鼠逃避潜伏期长于常规手术组,单位时间内穿越平台的次数、平台象限滞留的时间较常规手术组减少,差异有统计学意义(P<0.05)。各组大鼠脑海马组织中Srxnl、Nrf2蛋白表达水平比较,差异无统计学意义(P>0.05)。与对照组比较,假手术组脑脊液SOD活力值、Srxn1浓度差异有统计学意义(P<0.05)。结论:Srxn1的表达水平变化可能与VCI的发病关系不密切。Objective:To investigate the changes of thioredoxin-1(Srxn1)in the pathogenesis of vascular cognitive impairment(VCI)rat model.Methods:A total of 52 healthy male SD rats were randomly divided into 5 groups,6 rats in the control group were not given any treatment;6 rats in the sham operation group were only separated from the bilateral common carotid arteries;13 rats were in the conventional surgery group,and the bilateral common carotid arteries were ligated by the modified 2-V0 method;The bilateral common carotid arteries were ligated by the modified 2-V0 method,and the Srxn1 and Nrf2 gene interference fragments were injected into the lateral ventricle 24 hours before the operation to establish the VCI rat model;the spatial learning and memory ability of the rats in each group was evaluated according to the Morris water maze navigation test;the superoxide dismutase in the cerebrospinal fluid of each group was detected(SOD)activity,cerebrospinal fluid malondialdehyde(MDA)content,Srxn1 concentration,and protein expression levels of Srxn1 and Nrf2 in hippocampal tissue.Results:The escape latency of the rats in the conventional operation group was longer than that of the control group,and the times of crossing the platform per unit time and the time of staying in the quadrant of the platform were less than those of the control group,and the difference was statistically significant(P<0.05);On day 4 and 5,the escape latency of rats in Srxn1-siRNA surgery group and Nrf2-siRNA surgery group was longer than that in conventional surgery group,and the number of times of crossing the platform and the time of platform quadrant detention per unit time were reduced compared with that in conventional surgery group,with statistical significance(P<0.05).There was no significant difference in the expression levels of Srxn1 and Nrf2 proteins in the brain hippocampus of rats in each group(P>0.05).Compared with the control group,there were statistically significant differences in the SOD activity value and Srxn1 concentration in the
