摘　　要：目的:探讨WHAMM在食管鳞状细胞癌(ESCC)组织中的表达及对ESCC细胞生物学行为的影响。方法:收集22例ESCC患者手术切除的癌组织和癌旁正常食管黏膜组织标本,采用qRT-PCR检测WHAMM mRNA的表达。分别比较GSE67269(n=73)、GSE161533(n=28)数据集和本研究收集标本中ESCC组织和癌旁正常食管黏膜组织WHAMM mRNA的表达情况。Kaplan-Meier法绘制TCGA数据集中84例ESCC患者的生存曲线,采用Log-rank检验分析WHAMM的表达与患者预后的关系。取EC109和KYSE510细胞分为两组:过表达组细胞感染携带WHAMM过表达载体的慢病毒,阴性对照组感染携带空载体的对照病毒。采用CCK-8和Transwell法检测细胞的增殖、迁移和侵袭情况。结果:GSE67269、GSE161533数据集和本研究ESCC组织中WHAMM mRNA的表达均高于癌旁正常食管黏膜组织(P<0.05)。WHAMM低表达组生存状况好于高表达组(P=0.023)。与阴性对照组比较,过表达组EC109和KYSE510细胞的增殖、迁移和侵袭能力增强(P<0.05)。结论:WHAMM在ESCC中表达升高,可促进ESCC细胞的增殖、迁移和侵袭,与患者的预后相关。Aim:To investigate the expression of WHAMM in esophageal squamous cell carcinoma(ESCC)tissue and its effects on biological behaviors of ESCC cells.Methods:A total of 22 specimens of ESCC and normal mucosa tissue adjacent to the tumor after surgical resection were collected,and qRT-PCR was performed to detect the expression of WHAMM mRNA.The level of WHAMM mRNA in ESCC and adjacent normal esophageal mucosa tissue based on datasets GSE67269(n=73)and GSE161533(n=28)and clinical specimens of this study were compared.Kaplan-Meier method was employed to draw the survival curve of 84 ESCC patients from TCGA dataset.Log-rank analysis was conducted to analyze the relationship between the expression of WHAMM mRNA and the prognosis of the patients.EC109 and KYSE510 cells were divided into 2 groups,the overexpression group was transfected with lentiviruses overexpressing WHAMM,and the negative control(NC)group was transfected with lentiviruses carrying empty vector.CCK-8 assay and Transwell method were used to observe the proliferation,migration and invasion of the ESCC cells.Results:The WHAMM mRNA expression was up-regulated in ESCC tissue compared with adjacent normal esophageal mucosa tissue based on the clinical specimens and datasets GSE67269 and GSE161533(P<0.05).The ESCC patients with low WHAMM mRNA expression had better prognosis(P<0.05).EC109 and KYSE510 cells in the overexpression group exhibited enhanced proliferation,migration and invasion capabilities compared with NC group(P<0.05).Conclusion:The elevated expression of WHAMM can be observed in ESCC tissue which may prompt the proliferation,migration and invasion of ESCC cells,and correlate with the prognosis of the patients.
