摘 要:目的 了解重庆口岸首例输入性基孔肯雅病毒基因组特征和遗传变异情况,为口岸基孔肯雅热防控工作提供参考。方法 扩增病毒全基因组并测序,使用生物信息学软件对病毒株的全基因组遗传特征进行分析。结果 重庆口岸首例输入性基孔肯雅病毒命名为CHIK1801,序列全长11 672 nt,与参考序列(NC004162)核苷酸同源性为96.6%。结构基因包含3 747个核苷酸,编码1 248个氨基酸,与参考株S27(NC004162)相比有28个氨基酸变异位点。系统进化分析显示,与MN974219.1 (Thailand,2018)、MN974218.1 (Thailand,2018)、MN974211.1(Thailand,2018)、MK040571.1(Thailand,2018)、ON262796.1(Thailand,2019)、MT668625.1(Tianjin,2019)聚类在同一分支,均属东中南非洲遗传谱系(ECSA)印度洋分支。结论 重庆口岸首次检出的基孔肯雅病毒属东中南非洲遗传谱系(ECSA)印度洋分支,共有28个氨基酸位点变异,应加强基孔肯雅热监测。Objective To understand the genomic characteristics and genetic variation of the first imported chikungunya virus at Chongqing port,in order to provide reference for the prevention and control of chikungunya fever at ports.Methods The whole genome of the virus was amplified and sequenced,and the genetic characteristics of the whole genome was analyzed.Results The first imported chikungunya virus at Chongqing port was named CHIK1801,had a total length of 11672 nt.It shared 96.6%nucleotide homology with the reference sequence(NC004162).The structure protein gene contained 3747 nucleotides,encoding 1248 amino acids.Compared with the reference strain S27(NC004162),there were 28 amino acid mutation sites.Phylogenetic analysis showed that it clustered with MN974219.1(Thailand,2018),MN974218.1(Thailand,2018),MN974211.1(Thailand,2018),MK040571.1(Thailand,2018),0N262796.1(Thailand,2019),and MT668625.1(Tianjin,2019),and belonged to the Indian Ocean subtype of the East Central South African Genetic Pedigree(ECSA).Conclusion The first chikungunya virus detected at Chongqing port belonging to the East Central South Africa Genetic Genealogy(ECSA)Indian Ocean branch,with a total of 28 amino acid mutations.It is suggested to strengthen the monitoring of chikungunya fever.
