摘　　要：目的探讨长链非编码RNA LOC344887在非小细胞肺癌(NSCLC)组织中的表达及对NSCLC细胞生长的影响。方法利用ENCORI数据库中的基因芯片结果分析NSCLC组织和正常肺组织中LOC344887的表达水平。以肺鳞癌细胞系NCI-H2170为研究对象,采用干扰RNA法敲低细胞中LOC344887的表达,并采用实时荧光定量PCR法检测LOC344887的表达水平变化,克隆形成实验及CCK8法检测LOC344887对NCI-H2170细胞增殖的影响,流式细胞术分析LOC344887对NCI-H2170细胞周期的影响。最后利用TCGA数据库分析LOC344887表达水平与NSCLC患者总生存期的关系。结果LOC344887在NSCLC组织中尤其是肺鳞癌的表达显著高于正常肺组织(P<0.05)。转染siRNA后NCI-H2170细胞中LOC344887的表达显著下调(P<0.05),LOC344887敲低组的细胞增殖能力明显低于对照组(P<0.05),且细胞周期阻滞于G1期(P<0.05)。LOC344887与NSCLC患者的总生存期显著相关,LOC344887表达越高,患者总生存率越差(P<0.05)。结论LOC344887在肺鳞癌组织中高表达,影响细胞生长,且与患者预后相关,提示LOC344887可作为肺鳞癌的潜在治疗靶标和预后标志物。Objective To investigate the expression of long non-coding RNA LOC344887 in NSCLC tissues and its effect on the non-small cell lung cancer cell lines.Methods ENCORI database was used to analyze the expression of LOC344887 in NSCLC tissues and normal tissues.In this study,the expression of LOC344887 in lung squamous cell line NCI-H2170 was down-regulated by interference RNA,and the level of LOC344887 was detected by real-time fluorescence quantitative PCR.The proliferation of NCI-H2170 cells was detected by cloning formation assay and CCK8 assay,and cell cycle of NCI-H2170 was analyzed by flow cytometry.Additionally,TCGA database was used to analyze the relationship between the expression of LOC344887 and the overall survival of patients with NSCLC.Results The expression of LOC344887 in NSCLC tissues was significantly higher than that in normal tissues,especially in lung squamous cell carcinoma tissues(P<0.05).After transfection with siRNA,the level of LOC344887 in NCI-H2170 cells was downregulated markedly(P<0.05).The cell proliferation of LOC344887 knockdown group was significantly lower than that of the control group,and the cell cycle was arrested at G1 phase(P<0.05).LOC344887 was significantly associated with the overall survival of patients with NSCLC(P<0.05).The higher the expression of LOC344887,the worse the prognosis of patients(P<0.05).Conclusion LOC344887 affects the growth of lung squamous cell carcinoma cell NCI-2170.LOC344887 is highly expressed in NSCLC tissues and correlated with prognosis,suggesting that LOC344887 could be used as a potential therapeutic target and prognostic marker for lung squamous cell carcinoma.
