摘  要:LHCⅡ是围绕在光系统Ⅱ外部的捕光复合体,由Lhcb基因家族编码。为了解马尾松(Pinus massoniana)Lhcb基因(PmLhcb)家族特征,本研究利用转录组数据鉴定PmLhcb基因家族,并对其系统进化、结构和功能及不同叶型幼苗遮荫处理下的表达规律进行分析。结果显示,共鉴定出35个PmLhcb基因家族成员,在已报道的Lhcb 8个亚家族中,马尾松只包含Lhcb1、2、4、5和7这五个亚家族。本研究利用油松和买麻藤基因组数据进行Lhcb基因家族鉴定,得到相同的结果,表明Lhcb3、Lhcb6和Lhcb8亚家族为裸子植物和被子植物分化后才形成。生物信息学分析发现,PmLhcb蛋白主要定位在叶绿体,均含有叶绿素a/b结合结构域和跨膜结构,但Lhcb4亚家族的保守结构域存在较大的变异。实时荧光定量PCR分析表明,PmLhcb基因主要在马尾松幼苗的叶中大量表达,且Lhcb1和Lhcb2亚家族成员的表达量远高于其他亚家族;遮荫处理后PmLhcb基因在马尾松全初生叶幼苗和全次生针叶幼苗中上调表达,且在全次生针叶幼苗中上调的程度显著高于全初生叶幼苗。以上结果可为深入揭示PmLhcb基因家族功能、系统进化关系及其对光照的响应机制提供理论依据。LHCⅡis a light-harvesting complex that surrounds the photosystemⅡ,which is encoded by Lhcb gene family.In order to understand the characteristics of the Lhcb gene family of Pinus massoniana,the PmLhcb gene family was identified by transcriptome data,following by the analysis of phylogeny,structure,functions and expression pattern of seedlings with different leaf types under shading treatments.The results showed that 35 members of PmLhcb gene family were identified.Compare to the eight known subfamilies,P.massoniana only contained five subfamilies:Lhcb1,2,4,5 and 7.Similar results were obtained from the examination based on genomic data of P.tabulaeformis and Gnetum montanum.These evidences indicated that Lhcb3,Lhcb6 and Lhcb8 subfamilies were formed after the differentiation of gymnosperms and angiosperms.Bioinformatics analysis suggested that PmLhcb proteins were mainly located in chloroplast and contained chlorophyll a/b binding domain and transmembrane structure,but the conserved domain of Lhcb4 subfamily varied greatly.The results of real-time fluorescence quantitative PCR revealed that PmLhcb genes were mainly expressed in leaves,and the expressions of Lhcb1 and Lhcb2 subfamily members were much higher than that of the other subfamilies.After shading treatment,the expressions of PmLhcb genes were generally up-regulated in two seedling types,however the uptrends in secondary needles seedlings were significantly higher than that in primary needles seedlings.These researches could provide reference information for further studies of the function and phylogenetic relationship of Lhcb gene family.
