摘 要:目的通过网络药理学方法预测清肺合剂治疗非小细胞肺癌(NSCLC)的作用机制。方法检索中药系统药理学数据库与分析平台(TCMSP)、STITCH等相关数据库,筛选出清肺合剂10味中药所含活性成分及对应靶点;通过TCMSP、TTD、PharmGKB等数据库筛选出NSCLC相关靶点;将药物靶点与疾病靶点相交集得到清肺合剂治疗NSCLC的潜在作用靶点;通过Cytoscape 3.6.0软件构建“活性成分-潜在靶点”网络进行分析,筛选出关键活性成分;采用R包cluster Profiler对潜在靶点进行GO富集和KEGG通路分析。结果获得清肺合剂活性成分108个,药物靶点357个,清肺合剂治疗NSCLC的潜在作用靶点34个;网络分析显示清肺合剂治疗NSCLC关键活性成分包括槲皮素、木犀草素、熊果酸、芹菜素等,关键靶点包括90 kDa热休克蛋白αB1、转录因子p65、丝氨酸/苏氨酸蛋白激酶、血管内皮生长因子A等;GO富集和KEGG通路分析显示清肺合剂作用于NSCLC的途径包括PI3K-Akt信号通路、经典癌症通路、蛋白多糖、微小RNA等。结论该研究初步揭示了清肺合剂治疗NSCLC的多组分、多靶点、多途径的作用机制,为后续研究提供了参考和依据。Objective To predict the mechanism of action of Qingfei Mixture(清肺合剂)in the treatment of non-small cell lung cancer(NSCLC)by network pharmacology.Methods Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform TCMSP(TCMSP),STITCH and other related databases were retrieved to screen out the active ingredients and corresponding targets in 10 traditional Chinese drugs of Qingfei Mixture.NSCLC-related targets were screened through TCMSP,TTD,PharmGKB and other databases.The intersection of drug targets and disease targets were carried out to obtain potential targets of Qingfei Mixture in the treatment of NSCLC.The“active ingredient-potential target”network was constructed by Cytoscape 3.6.0 software for analysis,and the key active ingredients were screened out.GO enrichment and KEGG pathway analysis of potential targets were carried out by using the R package cluster Profiler.Results There were 108 active ingredients in Qingfei Mixture,357 drug targets and 34 potential targets of Qingfei Mixture in the treatment of NSCLC.Network analysis shows that the key active ingredients of Qingfei Mixture in the treatment of NSCLC include quercetin,luteolin,ursolic acid,apigenin,etc,and the key targets include HSP90AB1,RELA,AKT1,VEGFA,etc.GO enrichment and KEGG pathway analysis showed that the pathways that Qingfei Mixture acts on NSCLC include PI3K-Akt signaling pathway,classical cancer pathway,proteoglycan,microRNA,etc.Conclusion This study preliminarily revealed the multi-component,multi-target and multi-way mechanism of action of Qingfei Mixture in the treatment of NSCLC,and provided a reference and basis for subsequent research.
