摘  要:研究了灵芝水提物、灵芝多糖、灵芝乙醇提取物和灵芝乙酸乙酯相提取物对睡眠剥夺斑马鱼的睡眠改善作用,并探讨灵芝提取物对γ-氨基丁酸A型受体α1亚单位(GABRA1)、γ-氨基丁酸A型受体α3亚单位(GABRA3)、γ-氨基丁酸A型受体α5亚单位(GABRA5)、谷氨酸脱羧酶1(GAD1)、谷氨酸受体2A亚单位(GRIN2A)、5-羟色胺受体1A(HTR1A)、5-羟色胺受体2A(HTR2A)和褪黑素受体1类亚型(MTNR1A)mRNA表达的影响。研究结果表明4种灵芝提取物均具有睡眠改善效果。在睡眠改善效果上,灵芝水提物大于灵芝粗多糖,灵芝乙醇提取物大于灵芝乙酸乙酯相提取物。灵芝粗多糖能显著降低斑马鱼GABRA1,GRIN2A,GAD1 mRNA的表达,上调GABRA5,MTNR1A,HTR1A,HTR2A基因表达水平,从而改善斑马鱼的睡眠;灵芝乙酸乙酯相提取物能显著降低斑马鱼GAD1 mRNA的表达,上调GABRA3,GABRA5,GRIN2A,MTNR1A,HTR1A基因表达水平,从而缓解斑马鱼失眠模型的失眠效应。The objectives of this works were to study the effects of Ganoderma lucidum aqueous extract,Ganoderma lucidum crude polysaccharide,Ganoderma lucidum ethanol extract and Ganoderma lucidum acetate phase extract on sleep-depriving zebrafish,by accessing the mRNA levels of Ganoderma lucidum extracts on the mRNA expression ofγ-aminobutyric acid type A receptor alpha 1 subunit(GABRA1),γ-aminobutyric acid type A receptor alpha 3 subunit(GABRA3),γ-aminobutyric acid type A receptor alpha 5 subunit(GABRA5),glutamate decarboxylase 1(GAD1),glutamate receptor 2A subunit(GRIN2A),5-hydroxytryptamine receptor 1A(HTR1A),5-hydroxytryptamine receptor 2A(HTR2A)and melatonin receptor class 1 subtype(MTNR1A).The results presented that four Ganoderma lucidum extracts have sleep improvement effects.In terms of improvement,Ganoderma lucidum aqueous extract is greater than Ganoderma lucidum crude polysaccharide;Ganoderma lucidum ethanol extract is greater than Ganoderma lucidum acetate phase extract.Ganoderma lucidum crude polysaccharide significantly reduces mRNA expression of GABRA1,GRIN2A and GAD1 and upregulate the expression levels of GABRA5,MTNR1A,HTR1A and HTR2A genes,thus improving the sleep of zebrafish;ethyl acetate phase extract of Ganoderma lucidum significantly reduces the expression of its GAD1 mRNA and upregulate the expression levels of GABRA3,GABRA5,GRIN2A,MTNR1A and HTR1A genes,therefore alleviating the impact of sleeplessness in the zebrafish insomnia model.
