摘 要:目的基于网络药理学分析探讨槟榔促进口腔黏膜下纤维化(OSF)的活性成分以及其潜在的分子作用机制。方法利用中医药系统药理学数据库和分析平台(TCMSP)筛选槟榔的活性成分和相关潜在靶点;应用GeneCards和DisGeNET数据库建立OSF的基因靶点数据库;进一步采用Venny2.1.0数据处理平台和微生信数据处理平台取槟榔及OSF交集基因;基于槟榔潜在靶点与OSF靶点的匹配结果,构建蛋白质–蛋白质相互作用(PPI)网络,通过拓扑分析来筛选核心靶点;采用基于核心靶点的基因组(KEGG)通路富集分析及基因本体(GO)功能富集分析建立槟榔促进OSF的信号通路图。结果共筛选出槟榔5种活性成分,包括表儿茶素、原花青素B1等;213个槟榔相关的作用靶点以及596个OSF的疾病作用基因。通过取交集,获得31个关键相交基因,主要包括TP53、MDM2、MMP2等。GO功能富集分析显示,槟榔作用靶点共涉及到20条生物学过程,KEGG通路富集筛选出20条信号通路参与OSF的形成。结论初步证实了槟榔具有多成分-多靶点-多途径协同作用的特点以及促进OSF形成发病的潜在分子机制,为进一步探索其作用机制及临床应用奠定基础。【Objective】To explore the active components of areca nut in promoting oral submucous fibrosis(OSF)and its potential molecular mechanism based on network pharmacological analysis.【Methods】Using TCM systematic pharmacology database and analysis platform(TCMSP)to screen the active components and related potential targets of areca nut.Gene target database of oral submucous fibrosis was established by using GeneCards and DisGeNET databases.Further,the intersection genes of areca nut and oral submucous fibrosis were obtained by Venny 2.1.0 data processing platform and Micro Bioinformatics data processing platform.Based on the matching results of betel nut potential targets and oral submucous fibrosis targets,a PPI network was constructed to screen core targets through topology analysis.The core target based genome(KEGG)pathway enrichment analysis and gene ontology(GO)function enrichment analysis were used to establish the signal pathway map of areca nut promoting oral submucous fibrosis.【Results】A total of 5 active components of Areca catechu were screened,including epicatechin,procyanidin B1,etc;213 areca nut related targets and 596 oral submucous fibrosis disease genes.Through intersection,31 key intersection genes were obtained,mainly including TP53,MDM2,MMP2,etc.GO function enrichment analysis showed that areca nut target involved 20 biological processes,and KEGG pathway enrichment screened 20 signal pathways involved in the formation of oral submucous fibrosis.【Conclusion】It is preliminarily confirmed that areca nut has the characteristics of multi-component,multi target and multi pathway synergy and the potential molecular mechanism of promoting the formation of oral submucous fibrosis,which lays a foundation for further exploring its mechanism of action and clinical application.
