摘 要:建立了C_(18)-色谱柱串联亲水相互作用色谱(HILIC)柱高效测定咖啡饮料中咖啡因含量的方法。对色谱分离条件进行了优化,实验结果表明最佳的色谱分离条件为:甲醇/水=30∶70(V∶V),流速为0.5 mL/min,柱温为30℃,检测波长为272 nm。在4个咖啡饮料中均检出了咖啡因,咖啡饮料检出量为10.36~29.46 mg/L,相对标准偏差(RSD)不高于3%。检测下限(S/N=3)为0.1 mg/L。为验证方法的准确性,进一步开展了加标回收实验,4个咖啡饮料中咖啡因的回收率为81.08%~119.32%。该方法能满足实际样品中咖啡因的检测。并且双柱串联技术还消除了基质效应,该方法能准确、高效、灵敏地测定实际样品中的咖啡因。C_(18)-Hydrophilic interaction double-column tandem method was established to efficiently determine caffeine in coffee beverage.The chromatographic separation conditions were optimized and experimental results showed that the best conditions were mobile phase of methanol/water=30∶70(V∶V)with a flow rate of 0.5 mL/min,a column temperature of 30℃and a detection wavelength of 272 nm.Caffeine was detected in all four coffee beverages,the detected amount was 10.36~29.46 mg/L and the relative standard deviation(RSD)was below 3%.The lower detection limit(S/N=3)was 0.1 mg/L.In order to verify the accuracy of the method,further experiments were carried out to add standard recovery.The recoveries of 4 kind coffee beverages were between 81.08%and 119.32%.This method can satisfy the detection of caffeine in actual samples.In addition,the double-column tandem technology also eliminates the matrix effect so that this method can accurately,efficiently and sensitively determine the caffeine in actual samples.
