摘  要:目的了解广州市环境污水中沙门菌血清学分型、脉冲场凝胶电泳(pulsed-field gelelectrophoresis,PFGE)分子分型特征和耐药情况,为沙门菌引起的食源性疾病溯源和合理用药提供理论基础。方法2022年2月1日—2022年7月31日,对广州市环境污水进行定时定点采样,对收集到的污水进行增菌培养、分离纯化及生化鉴定。利用玻片凝集法对分离到的菌株进行血清学试验,用微量肉汤稀释法进行药敏实验,利用脉冲场凝胶电泳(PFGE)技术进行PFGE分子分型分析。结果共收集了288份污水,其中有66份污水检测出沙门菌,沙门菌检出率为22.92%。共分离出173株沙门菌,以鼠伤寒沙门菌、罗森沙门菌相对较多,另检出多种不常见血清型。共获得了97种PFGE图谱型,形成了2大聚类,同一血清型不同基因型和不同血清型同一基因型的情况都有出现。沙门菌对四环素、氨苄西林等抗生素高度耐药。多重耐药率高达87.50%,共有26种耐药谱。结论广州市污水中沙门菌血清型分布广泛,具有高度遗传多样性,且具有很大的传播耐药性风险,应加强沙门菌监测,并将环境污水监测当作常规监测。Objective To understand the serological typing,pulsed field gel electrophoresis(PFGE)molecular typing,and drug resistance of Salmonella in environmental sewage in Guangzhou,to provide theoretical basis for tracing the source of foodborne diseases caused by Salmonella and rational drug use.Methods From February 1,2022 to July 31,2022,the environmental sewage in Guangzhou was sampled at a fixed time point and site,and the collected sewage was cultured,isolated,and purified,followed by biochemical identification.The serological test of the isolated strains was carried out by slide agglutination method,the drug sensitivity test was carried out by microbroth dilution method,and the PFGE molecular typing was analyzed by PFGE.Results A total of 288 samples of sewage were collected,of which 66 samples were positive for Salmonella,and the detection rate of salmonella was 22.92%.A total of 173 strains of Salmonella were isolated,with relatively more Salmonella Typhimurium and Salmonella Rosanne,and a variety of unusual serotypes were detected.A total of 97 PFGE patterns were obtained,forming two clusters including the same serotype with different genotypes as well as different serotypes of the same genotype.Salmonella was highly resistant to tetracycline,ampicillin,and other antibiotics.The rate of multiple drug resistance was as high as 87.50%,and there were 26 kinds of drug resistance spectrum.Conclusion Salmonella serotype in sewage in Guangzhou is widely distributed and has high genetic diversity and a great risk of spreading drug resistance.Salmonella surveillance should be strengthened and environmental sewage monitoring should be regarded as routine monitoring.
