微米大黄炭对乙醇性急性胃黏膜损伤大鼠脂氧素A4合成途径的影响

摘  要:目的:研究微米大黄炭对乙醇诱导急性胃黏膜损伤模型大鼠黏膜的保护作用及脂氧素A4合成途径的影响。方法:将32只SD大鼠随机分为空白组、模型组、铝镁加混悬液组和微米大黄炭组,每组8只,各组分别予以相应浓度药物灌胃1 h后,采用无水乙醇法制备急性胃黏膜损伤大鼠模型,观察各组大鼠胃黏膜组织PGE_(2)、LXA_(4)、LTA_(4)、LTB_(4)水平,检测各组大鼠胃黏膜组织5-LOX、12-LOX、15-LOX及LXA4受体FPR2的蛋白表达水平。结果:各给药组大鼠黏膜损伤指数显著低于模型组。与模型组相比,铝镁加组PGE2、LXA_(4)、15-LOX表达水平显著上升,LTA_(4)、LTB_(4)表达水平均显著下降,5-LOX、12-LOX、FPR2表达水平差异无统计学意义;微米大黄炭组PGE_(2)、LXA_(4)、5-LOX、15-LOX、FPR2表达水平显著上升,LTA_(4)、LTB_(4)表达水平显著下降,12-LOX表达水平差异无统计学意义;组间比较,微米大黄炭组较铝镁加组PGE_(2)、LXA_(4)、5-LOX、15-LOX、FPR2表达水平升高更为显著,LTA_(4)、LTB_(4)表达水平差异无统计学意义。结论:微米大黄炭对乙醇诱导急性胃黏膜损伤大鼠的保护作用机制可能通过上调PGE2、FPR2表达水平,激活15/5-LOX催化LXA4合成途径,发挥促进黏膜修复作用。OBJECTIVE To study the protective effect of micronized rhubarb charcoal on ethanol induced acute gastric mucosa injury in rats and the influence of lipoxygen-A synthesis pathway.METHODS Thirty-two SD rats were equally randomized into four groups as blank,model,almagate suspension(AS),and micron rhubarb charcoal(MRC).After pretreated with intragastric administration of drugs in corresponding concentrations,rats were subjected to intragastric administration of ethanol for causing the mucosal damage.Mucosal concentrations of PGE_(2),LXA_(4),LTA_(4),LTB_(4) and expressions of 5-LOX,12-LOX,15-LOX,and LXA4 receptor FPR2 in gastric mucosa were measured.RESULTS The mucosal injury index(UI)of drug administered rats was significantly lower than that in the model group.Significant increase in the expression levels of PGE_(2),LXA_(4),15-LOX and decline in LTA4 and LTB4 were observed in AS group compared with the model group.5-LOX,12-LOX,and FPR2 levels were non-statistically different;the micron rhubarb charcoal group had significantly increased expression levels of PGE_(2),LXA_(4),5-LOX,15-LOX,and FPR2 expression levels,LTA4 and LTB4 expression levels were significantly decreased,and 12-LOX expression level was not statistically different.The micron rhubarb charcoal was observed even more significant in elevating the levels of PGE_(2),LXA_(4),5-LOX,15-LOX,FPR2 than AS did,while no statistical difference was observed in the LTA4 and LTB4 expressions.CONCLUSION Micron rhubarb charcoal may exert its gastroprotective effect against ethanol induced acute gastric mucosal injury via promoting mucosal repairment by upregulating the PGE2 and FPR2 expressions and activating the 15/5-LOX-catalyzed LXA4 synthesis pathway.
