摘 要:目的本研究分析前列腺癌(PCa)患者外周血DNA腺嘌呤N6甲基化(6mA)修饰的水平,从而探讨6m A修饰与PCa的关系,为PCa寻找新的诊断和预后标志物。方法分别检测PCa组与对照组的外周血6mA,筛选差异基因6mA热点,通过生物信息学分析方法,在全基因组范围对疾病相关位点进行筛选与鉴定。结果与正常组比较,PCa组中6mA表达上调基因有2558个,表达下调基因有1106个,主要分布在内含子上。功能富集与信号通路富集分析结果显示,钙离子信号通路和T细胞受体信号通路富集倍数最高,此通路中PLCG1、ITPR1、PPP3C和CAMK2等上调基因与上游T细胞受体调控基因共同作用调控钙离子信号转导。结论钙离子信号通路和T细胞受体信号通路6m A修饰异常可能与PCa的发生、发展相关。Objective We analyzed the level of DNA adenine N6 methylation(6mA)in peripheral blood from patients with prostate cancer(PCa),so as to explore the relationship between 6mA and PCa,and to search for new diagnostic and prognostic markers for PCa.Methods Peripheral blood was isolated from PCa and negative control(NC)groups,and was sentenced to screen differential genes with 6mA peak.Through bioinformatics analysis,disease-related loci was identified by genome-wide study.Results There were 2558 up-regulated 6mA genes and 1106 down-regulated 6mA genes in PCa group,mainly distributed in introns.Using GO enrichment and KEGG signaling pathway analysis,the differential 6mA-associated genes were mainly enriched in calcium signaling pathway and T cell receptor signaling pathway.In this pathway,up-regulated genes such as PLCG1,ITPR1,PPP3C,CAMK2 interact with upstream T cell receptor regulatory genes to regulate calcium signal transduction.Conclusions Abnormal modification of calcium signaling pathway and T cell receptor signaling pathway 6mA may be closely related to the occurrence and development of PCa.
