摘 要:目的研究细胞周期蛋白B1(cyclin B1,CCNB1)在肝细胞癌(hepatocellular carcinoma,HCC)组织中的表达及其临床研究。方法从基因表达综合数据库(GEO)获得4个HCC相关数据集,分析HCC和正常肝组织之间的差异表达基因。从癌症基因组图谱(TCGA)数据库获得374例HCC患者的临床数据及基因表达谱,验证CCNB1的表达水平。利用人类蛋白图谱(HPA)数据库分析CCNB1的蛋白表达水平。Kaplan-Meier plotter数据库评估CCNB1对肝癌患者的预后价值。用单因素及多因素Cox回归分析影响HCC患者总生存期(OS)的危险因素。绘制受试者工作特征(ROC)曲线,并计算曲线下面积(AUC)评估CCNB1表达的诊断价值。结果CCNB1在HCC组织中高表达。生存分析显示CCNB1的高水平表达与HCC患者的OS成负相关(HR=2.34,95%CI:1.55~3.54,P<0.001)。多因素Cox回归分析显示,CCNB1高表达是影响HCC患者OS的危险因素(HR=1.991,95%CI:1.252~3.168,P=0.004)。ROC曲线分析显示,CCNB1的AUC为0.980,具有较高的诊断价值。结论CCNB1在HCC组织中高表达,并与HCC患者的预后不良相关,CCNB1可能是HCC患者诊断和预后评估的有价值的生物学指标。Objective This paper aims to investigate the expression and its potential clinical significance of cyclin B1(CCNB1)in Hepatocellular carcinoma(HCC).Methods Four HCC-related datasets were obtained from the Gene Expression Omnibus Database(GEO)to analyze the differentially expressed genes between HCC and normal liver tissue.Clinical data and gene expression profiles of 374 HCC patients were obtained from the Cancer Genome Atlas(TCGA)database to verify the expression of CCNB1.The protein level of CCNB1 was analyzed using the Human Protein Atlas(HPA)database.Kaplan-Meier plotter database was adopted to evaluate the prognostic value of CCNB1 in patients with liver cancer.Univariate and multifactorial Cox regression were used to analyze the risk factors affecting overall survival(OS)of HCC patients.Receiver operating characteristic(ROC)curves were plotted and area under the curve(AUC)was calculated to evaluate the diagnostic value of CCNB1 expression.Results CCNB1 is highly expressed in HCC tissues.Survival analysis showed that high level of CCNB1 expression was negatively correlated with OS in HCC patients(HR=2.34,95%CI:1.55-3.54,P<0.001).Multivariate Cox regression analysis showed that high expression of CCNB1 was a risk factor for OS in HCC patients(HR=1.991,95%CI:1.252-3.168,P=0.004).ROC curve analysis showed that the AUC under the curve of CCNB1 was 0.980,which had high diagnostic value.Conclusion CCNB1 is highly expressed in HCC tissues and is associated with poor prognosis of HCC patients.CCNB1 may be a valuable biological indicator for diagnosis and prognosis evaluation of HCC patients.
