摘 要:以龙眼参为研究对象,采用响应面法优化超声辅助低共熔溶剂提取龙眼参黄酮工艺,并进一步探索其抗运动疲劳的功效。考察低共熔溶剂的提取效率,且经单因素试验和响应面试验选出最优提取条件,结果表明提取龙眼参黄酮的最佳工艺:含水量30%、摩尔比1∶3的氯化胆碱-乳酸为提取液、超声提取功率为156 W、提取温度为68℃、提取耗时为42 min和料液比为1∶30 g/mL,在最优提取条件下龙眼参黄酮提取率为37.98±0.13 mg/g。使用小鼠负重游泳模型进行抗运动疲劳的研究,结果表明龙眼参黄酮显著延长小鼠的力竭时间,降低运动后小鼠的血乳酸(BLA)、血尿素氮(BUN)和丙二醛(MDA)指标并增加其肝糖原(HG)和肌糖原(MG)的储备量,从而提升抗运动疲劳能力。The extraction process of flavonoids from Longan ginseng using ultrasonic-assisted deep eutectic solvents was optimized by response surface methodology,and anti-exercise fatigue effect of the flavonoids was investigated.The extraction efficiency of deep eutectic solvent and the optimum extraction conditions were studied by single factor experiment and response surface methodology.The results showed that the optimum extraction conditions were as follows:choline chloride-lactic acid with 30% water and molar ratio of 1∶3 as the extraction solution,ultrasonic extraction power 156 W,extraction temperature 68℃,extraction time 42 min and solid-liquid ratio 1∶30 g/mL.Under these conditions,the extraction rate of flavonoids was 37.98±0.13 mg/g.A mouse weight-loaded swimming model was used for anti-exercise fatigue study.The results showed that flavonoids from Longan ginseng significantly prolonged the exhaustion time of mice,reduced the indexes of BLA,BUN and MDA of mice after exercise,and increased the amount of HG and MG and improved anti-exercise-fatigue ability of mice.
