摘  要:目的通通过调节癌症相关成纤维细胞(CAF)的自噬水平来探究其对前列腺癌细胞的作用。方法分别检测正常成纤维细胞(NF)与CAF的自噬水平,随之使用氯喹抑制CAF的自噬水平,通过检测LC3I进一步确认CAF的自噬水平是否被抑制,然后分别与前列腺癌细胞PC-3和DU-145共培养,通过CCK-8增殖与Transwell侵袭实验分别评估前列腺癌细胞的增殖及侵袭能力。通过上调自噬起始基因Beclin-1来上调CAF的自噬水平,然后再次建立CAF与前列腺癌细胞共培养体系,评估前列腺癌细胞的增殖及侵袭能力;最后检测代表前列腺癌细胞侵袭能力的相关蛋白(MMP-9和MMP-2)的表达水平。结果CAF的自噬水平高于NF(P<0.05);与自噬抑制后的CAF共培养的前列腺癌细胞的增殖、侵袭能力明显降低(P<0.05);与自噬上调后的CAF共培养的前列腺癌细胞的增殖、侵袭能力明显增高(P<0.05);免疫印迹结果显示前列腺癌细胞中代表侵袭能力的相关蛋白表达水平增高(P<0.05)。结论仑CAF自噬水平高于NF的自噬水平;CAF的自噬可促进前列腺癌细胞增殖和侵袭。Objective To investigate the effect of Cancer associated fibroblasts(CAF)on prostate cancer cells by regulating the level of autophagy of CAF.Methods Autophagy levels of normal fibroblasts(NF)and CAF were detected respectively,and then chloroquine was used to inhibit the autophagy level of CAF,and whether the autophagy level of CAF was further confirmed by detecting LC3 I,and then co-cultured with prostate cancer cells PC-3 and DU-145 respectively.The proliferation and invasion ability of prostate cancer cells were evaluated by CCK-8 proliferation and Transwell invasion assay respectively.The autophagy level of CAF was up-regulated by up-regulating the autophagy initiation gene Beclin-l,and then the co-culture system of CAF and prostate cancer cells was established again to evaluate the proliferation and invasion ability of prostate cancer cells.Finally,the expression levels of related proteins(MMP-9 and MMP-2),which represent the invasion ability of prostate cancer cells,were detected.Results The autophagy level of CAF was higher than that of NF(P<0.05).The proliferation and invasion ability of prostate cancer cells co-cultured with CAF inhibited by autophagy were significantly decreased(P<0.05).The proliferation and invasion ability of prostate cancer cells co-cultured with CAF after autophagy were significantly increased(P<0.05).Western blot results showed that the expression level of related proteins representing invasion ability was increased in prostate cancer cells(P<0.05).Conclusions The autophagy level of CAF was higher than that of NF.Autophagy of CAF can promote the proliferation and invasion of prostate cancer cells.
