摘 要:近年来,代谢综合征发病率不断上升,药物治疗存在一定局限性,通过天然植物干预代谢综合征发生发展已成为研究热点。该研究通过果糖诱导的大鼠代谢综合征模型和网络药理学探讨青钱柳水提物调节糖脂代谢紊乱的作用及机制。实验设置正常对照组、代谢综合征模型组、青钱柳干预组、青钱柳对照组。代谢综合征模型组和青钱柳干预组通过10%果糖溶液长期替代饮用水诱导代谢综合征。青钱柳干预组和青钱柳对照组每天用青钱柳水提物(800 mg/kg)灌胃,模型组用等体积生理盐水灌胃。动态观察大鼠基本状态、体质量、空腹血糖值。10周后测量大鼠口服葡萄糖耐量、胰岛素耐量、腰围、体长、肝重,计算Lee′s指数、肝脏指数,对肝脏HE染色并评分,检测血清和肝脏组织总胆固醇、甘油三酯、低密度脂蛋白、高密度脂蛋白、肿瘤坏死因子-α、白细胞介素-1β、白细胞介素-6水平、超氧化物歧化酶、谷胱甘肽过氧化物酶、丙二醛、晚期糖基化终末产物水平。基于文献及数据库筛选重要靶点,构建“青钱柳-成分-靶点-代谢综合征”网络及蛋白-蛋白互作网络,并完成KEGG通路富集分析。实验结果表明,青钱柳水提物可显著改善果糖诱导的大鼠体重、腰围、Lee′s指数、肝脏指数及病理变化,调节糖脂代谢紊乱,缓解肝脏脂质过氧化及氧化损伤,降低血清及肝脏炎症因子水平。网络药理学结果提示其糖脂代谢调节的物质基础主要为酚类物质,机制涉及炎症和氧化应激,重要信号通路为AGE-RAGE等,与动物实验结果相印证。综上,青钱柳水提物对果糖诱导的代谢综合征大鼠糖脂代谢紊乱具有积极的调节作用,机制涉及炎症及氧化应激。The incidence of metabolic syndrome has been increasing,and there are limitations in traditional drug therapy,so intervention in the development of metabolic syndrome through natural products has become a hot research topic.This study investigated the role and mechanism of the aqueous extract of Cyclocarya paliurus in regulating disorders of glucolipid metabolism through a fructose-induced metabolic syndrome model and network pharmacology in rats.The experiment was set up with a normal control group,metabolic syndrome model group,Cyclocarya intervention group,and Cyclocarya control group.The metabolic syndrome was induced by a 10%fructose solution instead of drinking water.The Cyclocarya intervention group and Cyclocarya control group were given an aqueous extract of Cyclocarya at a dose of 800 mg/(kg·d),and the model group was given saline gavage at the same volume.The rats were dynamically observed for basic morphology,body mass,and fasting blood glucose.After 10 weeks,oral glucose tolerance test,insulin tolerance test,waist circumference,body length,and liver weight of rats were measured,Lee′s index and liver index were calculated,HE staining of the liver was performed and scored,and serum and liver tissue levels of triglyceride,total cholesterol,low-density lipoprotein,high-density lipoprotein,tumor necrosis factor-α,interleukin-1β,interleukin-6,superoxide dismutase,glutathione peroxidase,malondialdehyde,and advanced glycation end products levels.Based on the literature and database,this study screened the important targets and constructed the network of“cyanidin-component-target-metabolic syndrome”,protein-protein interaction network,and enrichment analysis of KEGG.The experimental results showed that the aqueous extract of Cyclocarya could significantly improve the body weight,waist circumference,Lee′s index,liver index,and pathological changes induced by fructose,regulate the disorders of glucolipid metabolism,alleviate liver lipid peroxidation,and oxidative damage,and reduce the levels of serum
