摘 要:优化坚果中总氰的前处理条件并建立连续流动分析仪检测总氰的方法。以0.30 mol/L NaOH溶液、70℃超声60 min作为最佳前处理方法浸提坚果中总氰,通过连续流动仪在600 nm下进行定量测定。在该方法中,总氰的检出限为0.05 mg/kg,定量限为0.15 mg/kg。在0.2~100μg/L范围内线性关系良好,r=0.9995;对杏仁样品做2.50,7.50和12.50 mg/kg三个水平总氰添加试验,加标回收率为92.4%~108.7%,SRSD为1.3%~2.0%(n=6)。该方法对实际样品测定与GB 5009.36—2016《食品安全国家标准食品中氰化物的测定》第一法分光光度法测定结果相比较,Z=-0.105,P=0.917>0.05,结果差异无统计学意义。该方法简化了传统方法的提取及检测操作程序,自动化程度高,减少试剂用量,且灵敏度高、精密度、准确度好,可作为大批量食品样品中总氰的检测方法之一。Optimize the pretreatment conditions of total cyanide in nuts,and establish a continuous flow analyzer method for the determination of total cyanide.0.30 mol/L NaOH solution and 70℃ultrasound for 60 min were used as the optimal pretreatment method to extract total cyanide from nuts.The total cyanide was quantified at 600 nm by continuous flow meter.The detection limit of total cyanide was 0.05 mg/kg,and the quantification limit was 0.15 mg/kg.The linearity between 0.2μg/L and 100μg/L was good,and r was 0.9995.Total cyanide at three levels of 2.50,7.50 and 12.50 mg/kg was added to almond samples,the recovery rate was 92.4%to 108.7%and the SRSD was 1.3%to 2.0%(n=6).The results of this method were compared with the results of the first method in GB 5009.36—2016 National Food Safety Standard Determination of Cyanide in Food,Z was-0.105,P was 0.917>0.05,and there was no significant difference in the results.This method simplified the extraction and detection procedure of the traditional method,had high degree of automation,reduced the amount of reagent,had high sensitivity,precision and accuracy,and could be used as one of the methods for the detection of total cyanide in large quantities of food samples.
