摘 要:目的:探讨枸杞多糖(LBP)对梗阻性黄疸(OJ)小鼠肝损伤的改善及保护作用。方法:本研究采用随机对照试验设计,共纳入成年健康雄性昆明小鼠60只,采用随机数字表法分为3组,即假手术组、模型组和枸杞组;假手术组只进行腹部切口手术,不进行模型构建和LBP干预,模型组和枸杞组行胆总管结扎致肝损伤构建OJ模型,枸杞组于手术后2d行固定时间LBP灌胃;6周后取材制样,行血清学检测[总胆红素(TBIL)、谷丙转氨酶(ALT)、谷草转氨酶(AST)]和免疫组化分析[肿瘤坏死因子α(TNF-α)、核转录因子κB(NF-κB)、IκB激酶α(IKK-α)、NF-κB抑制因子α(IκB-α)],评价LBP干预对OJ小鼠肝功能的保护作用。结果:模型组小鼠血清TBIL、ALT、AST水平处于三组的最高水平,枸杞组的TBIL、ALT、AST水平较模型组有明显降低,差异均有统计学意义(均P<0.001);但枸杞组的TBIL、ALT、AST水平仍然分别高于假手术组,差异均有统计学意义(均P<0.001)。模型组小鼠肝组织中TNF-α、NF-κB、IKK-α的表达处于三组的最高水平,枸杞组的TNF-α、NF-κB、IKK-α表达水平较模型组有明显下调,差异均有统计学意义(均P<0.001);但枸杞组的TNF-α、NF-κB、IKK-α表达水平仍然分别高于假手术组,差异均有统计学意义(均P<0.001);而IκB-α的表达水平则呈完全相反的趋势,差异均有统计学意义(均P<0.05)。结论:LBP干预对OJ小鼠的肝功能具有保护作用,可能通过抑制肿瘤坏死因子α介导的核转录因子κB信号通路发挥作用。Objective:To investigate the improvement and protective effect of lycium barbarum polysaccharide(LBP)on liver injury in obstructive jaundice(OJ)rats.Methods:A total of 60 adult healthy male Kunming rats were included in this study.They were randomly divided into three groups:sham operation group,model group and lycium barbarum polysaccharide group.The sham operation group only underwent abdominal incision operation without model construction and LBP intervention.The model group and lycium barbarum polysaccharide group were treated with common bile duct ligation to cause liver injury to construct OJ model.The lycium barbarum polysaccharide group was treated with LBP for a fixed time 2 days after operation.After 6 weeks,samples were taken and prepared for serological detection(TBIL,ALT,AST)and immunohistochemical analysis(TNF-α,NF-κB,IKK-α,IκB-α),to evaluate the protective effect of LBP intervention on liver function in OJ rats.Results:The levels of serum TBIL,ALT and AST in the model group were the highest in the three groups.The levels of TBIL,ALT and AST in the lycium barbarum polysaccharide group were significantly lower than those in the model group(all P<0.001);however,the levels of TBIL,ALT and AST in lycium barbarum polysaccharide group were still higher than those in sham operation group(all P<0.001).The expression of TNF-α,NF-κB,IKK-αin liver tissue of model group rats was the highest in the three groups.The expression level of TNF-α,NF-κB,IKK-αin lycium barbarum polysaccharide group was significantly lower than that in the model group(all P<0.001);but the expression level of TNF-α,NF-κB,IKK-αin lycium barbarum polysaccharide group was still higher than that in the sham operation group(all P<0.001),and the expression level of IκB-αshowed a completely opposite trend,and the differences were statistically significant(all P<0.05).Conclusion:LBP intervention has a protective effect on the liver function of OJ rats,possibly by inhibiting TNF-αmediated NF-κB signal pathway plays a role.
