摘 要:目的:探讨刺五加多糖(ASPS)对骨肉瘤小鼠的抑瘤作用及对免疫功能的影响。方法:通过前肢后内侧注射MG63细胞悬液建立骨肉瘤小鼠模型;将造模后的小鼠随机分为模型组、刺五加多糖低、中、高(36.25、72.5、145 mg/kg)剂量组、顺铂组(25 mg/kg顺铂),并以正常小鼠为对照组,每组10只;干预结束后,ELISA检测血清中白介素-2(IL-2)、干扰素-γ(IFN-γ)以及肿瘤坏死因子-α(TNF-α)水平;流式细胞仪检测外周血中CD4^(+)、CD8^(+)细胞比例;分离胸腺及脾脏,计算胸腺指数和脾脏指数;分离肿瘤,称量瘤体重量并计算抑瘤率;HE检测肿瘤组织病理学变化;Western blot检测肿瘤组织程序性死亡因子1(PD-1)及配体(PD-L1)蛋白表达。结果:与对照组相比,小鼠血清中IL-2、IFN-γ、TNF-α水平、外周血中CD8^(+)、瘤体重量、肿瘤组织中PD-1、PD-L1蛋白表达显著增加,脾脏指数、胸腺指数、CD4^(+)及CD4^(+)/CD8^(+)显著降低(P<0.05);与模型组相比,刺五加多糖低、中、高剂量组部分细胞核分裂,肿瘤细胞减少,血清中IL-2、IFN-γ、TNF-α水平、外周血中CD8^(+)、瘤体重量、肿瘤组织中PD-1、PD-L1蛋白表达均显著降低,脾脏指数、胸腺指数、CD4^(+)及CD4^(+)/CD8^(+)、抑瘤率均显著增加(P<0.05),但顺铂组与刺五加多糖高剂量组比较差异无统计学意义(P>0.05)。结论:ASPS可抑制骨肉瘤小鼠肿瘤生长,改善小鼠免疫功能。Objective:To investigate the anti-tumor effect of acanthopanax senticosus polysaccharides(ASPS)on osteosarcoma mice and its impact on immune function.Methods:A mouse model of osteosarcoma was established by injecting MG63 cell suspension into the posterior medial side of the forelimbs;the model mice were randomly divided into model group,Acanthopanax senticosus polysaccharide low(36.25 mg/kg),medium(72.5 mg/kg),high(145 mg/kg)dose groups,and cisplatin group(25 mg/kg cisplatin),with normal mice as the control group,with 10 mice in each group;after the intervention,ELISA was applied to detect serum levels of IL-2,IFN-γ,and TNF-α;flow cytometry was applied to detect the proportions of CD4^(+)and CD8^(+)cells in peripheral blood;the thymus and spleen were separated,and the thymus index and spleen index were calculated;the tumor was separated,the tumor mass was weighed,and the tumor inhibition rate was calculated;HE was applied to detect histopathology changes of tumors;Western blot was applied to detect the expression of programmed death factor-1(PD-1)and ligand(PD-L1)proteins in tumor tissue.Results:Compared with the control group,the levels of IL-2,IFN-γ,TNF-αin mouse serum,CD8^(+)in peripheral blood,tumor mass,and the expression of PD-1 and PD-L1 proteins in tumor tissue were obviously increased,the spleen index,thymus index,CD4^(+)and CD4^(+)/CD8+were obviously decreased(P<0.05).Compared with the model group,the Acanthopanax senticosus polysaccharide low-dose group,medium-dose group,high-dose group showed partial nuclear division and reduced tumor cells,the levels of IL-2,IFN-γ,TNF-α in serum,CD8^(+)in peripheral blood,tumor mass,and the expression of PD-1 and PD-L1 proteins in tumor tissue were obviously decreased,the spleen index,thymus index,CD4^(+),CD4^(+)/CD8^(+),and tumor inhibition rate were obviously decreased(P<0.05).However,there was no obvious difference between the cisplatin group and the Acanthopanax senticosus polysaccharide high-dose group(P>0.05).Conclusion:ASPS can inhibit tumor growth and
