摘　　要：目的 探讨脯氨酰4-羟化酶α亚单位3(P4HA3)在胃癌中的表达及临床意义。方法 通过UALCAN数据库及基因表达综合(GEO)数据库分析胃癌组织和癌旁正常组织中P4HA3基因表达差异,并进一步分析癌症基因组图谱(TCGA)数据库中不同临床分期、分化程度、年龄和性别胃癌患者P4HA3基因表达差异。以P4HA3基因表达中位值为界,将患者分为P4HA3高表达组和P4HA3低表达组,采用Kaplan-Meier plotter在线数据库比较两组患者的总生存时间、首次进展生存时间和进展后生存时间。收集107例胃癌患者的胃癌组织及癌旁正常组织标本,采用免疫组织化学染色法检测P4HA3蛋白表达情况,并结合临床特征及随访资料进行综合分析。结果 TCGA数据库和GEO数据库中的数据均显示,胃癌组织中P4HA3基因表达水平明显高于癌旁正常组织(P﹤0.01)。TCGA数据库显示,不同年龄、分化程度、临床分期胃癌患者的P4HA3基因表达水平比较,差异均有统计学意义(P﹤0.05)。Kaplan-Meier plotter在线数据库分析发现,P4HA3高表达组患者的中位总生存时间、中位首次进展生存时间、中位进展后生存时间均明显短于P4HA3低表达组,差异均有统计学意义(P﹤0.01)。胃癌组织中P4HA3蛋白阳性表达率高于癌旁正常组织(P﹤0.05)。不同肿瘤直径、分化程度、TNM分期、T分期胃癌患者胃癌组织中P4HA3蛋白表达情况比较,差异均有统计学意义(P﹤0.05);P4HA3阴性表达患者的累积生存率明显高于P4HA3阳性表达患者(P﹤0.01)。结论 P4HA3在胃癌组织中高表达,且与胃癌的发生发展及预后有关,可能成为胃癌的预后评估指标和潜在治疗靶点。Objective To investigate the expression of prolyl 4-hydroxylase subunit alpha 3(P4HA3)in gastric can-cer and its clinical significance.Method The differences of P4HA3 gene expression in gastric cancer tissues and adja-cent normal tissues were analyzed by the UALCAN database and the Gene Expression Omnibus(GEO)database,and P4HA3 gene expression differences in gastric cancer patients with different clinical stages,differentiation degrees,ages,and genders in The Cancer Genome Atlas(TCGA)database were further analyzed.The patients were divided into P4HA3 high-expression group and P4HA3 low-expression group based on the average value of the P4HA3 gene expression.The Kaplan-Meier plotter online database was used to compare the overall survival time,first-progression survival time,and post-progression survival time between the two groups.Gastric cancer tissue and adjacent normal tissue samples from 107 patients with gastric cancer were collected.The expression of P4HA3 protein was detected by immunohistochemical staining,and a comprehensive analysis was carried out in combination with clinical characteristics and follow-up data.Result The data in both TCGA database and the GEO database showed that the expression level of the P4HA3 gene in gastric cancer tissues was significantly higher than that in adjacent normal tissues(P<0.01).The TCGA database showed that there were statistically significant differences in the expression levels of the P4HA3 gene in gastric cancer patients with different ages,differentiation degrees,and clinical stages(P<0.05).Kaplan-Meier plotter online database analysis showed that the median overall survival time,median first-progression survival time,and median post-progression surviv-al time in the P4HA3 high-expression group were significantly shorter than those in the P4HA3 low-expression group,and the differences were statistically significant(P<0.01).The positive expression rate of P4HA3 protein in gastric cancer tissues was higher than that in adjacent normal tissues(P<0.05).There were stat
