摘 要:目的通过用不同强度和频率的电流刺激牛蛙离体坐骨神经-腓肠肌标本,了解复方当归注射液对腓肠肌收缩张力、收缩形式和兴奋性的影响。方法用双毁髓法制备坐骨神经-腓肠肌离体标本,在BL-420F生物信息采集系统中,记录滴加0.2 mL复方当归注射液10 min后,腓肠肌收缩波形的变化。结果复方当归注射液可分别降低坐骨神经干产生动作电位和腓肠肌单收缩时的阈强度、半高刺激强度、最适刺激强度(P<0.05或P<0.01);增强其兴奋性,增加单收缩时的最大、最小收缩张力和平均张力(P<0.05或P<0.01);同时能降低腓肠肌强直收缩时的最低刺激频率,增强其收缩时的最大、最小张力,且差异具有统计学意义(P<0.05或P<0.01)。维拉帕米和琥珀胆碱可阻断其对腓肠肌单收缩活动的上述作用(P<0.05),乙酰胆碱能增强其对腓肠肌单收缩时阈强度和最适刺激强度的作用(P<0.05)。结论复方当归注射液可促进坐骨神经干动作电位的传导,提高腓肠肌收缩兴奋性,其机制可能与促进钙离子内流、增加坐骨神经末梢乙酰胆碱(Ach)释放、促进Ach与腓肠肌细胞膜N2受体结合有关。Objective To understand the effect of compound angelica sinensis injection on the contraction tension,contraction form and excitability of the isolated bullfrog sciatic nerve-gastrocnemius muscle specimen by stimulating the current with different intensity and frequency.Methods The isolated specimens of the sciatic nerve-gastrocnemius muscle were prepared by the double pulp destruction method.In the BL-42F biological information collection system,the changes of the contraction waveform of the gastrocnemius muscle were recorded after 10 minutes of dripping 0.2 mL compound angelica sinensis injection.Results Compound angelica sinensis injection could reduce the threshold intensity,semi-high stimulation intensity and optimal stimulation intensity of action potential produced by sciatic nerve trunk and single contraction of gastrocnemius muscle respectively(P<0.05 or P<0.01);Enhance its excitability,increase the maximum,minimum,and average contractile tension during single contraction(P<0.05 or P<0.01);At the s-ame time,it can reduce the minimum frequency of stimulation during the contraction of gastrocnemius muscle rigidity,and enhance the maximum and minimum tension during the contraction,and the difference is statistically significant(P<0.05 or P<0.01).Verapa-mil and Suxamethonium chloride could block the above effects on the single contraction activity of gastrocnemius muscle(P<0.05),and Acetylcholine could enhance its effects on the threshold intensity and the optimal stimulus intensity of gastrocnemius muscle single contraction(P<0.05).Conclusion Compound angelica sinensis injection can promote the conduction of the Action potential of the sciatic nerve stem and increase the excitability of gastrocnemius muscle contraction.The mechanism may be related to promoting the influx of calcium ions,increasing the release of Acetylcholine(Ach)from the sciatic free nerve ending,and promoting the binding of Ach to the N2 receptor of the gastrocnemius muscle cell membrane.
