摘 要:本试验旨在通过研究不同年龄牦牛瘤胃、网胃、瓣胃、皱胃中表皮生长因子(EGF)与表皮生长因子受体(EGFR)的分布及表达差异,以探索EGF、EGFR与牦牛胃组织结构及生理功能之间的关系。本试验分别选取初生组、幼龄组、成年组和老年组健康牦牛,每组3头,屠宰后取其新鲜组织及分子样品;利用实时荧光定量PCR、免疫组织化学、免疫荧光技术,对EGF和EGFR在牦牛瘤胃、网胃、瓣胃、皱胃中的表达与分布进行研究。结果显示,EGF和EGFR主要在四个年龄段牦牛前胃(瘤胃、网胃、瓣胃)的黏膜上皮层中呈阳性分布,上皮细胞中阳性产物较多;皱胃中胃底腺区壁细胞和颈黏液细胞中呈阳性分布,且不同年龄组表达水平存在显著差异(P<0.05);EGF和EGFR在老年组各胃中相对表达量最低,初生组瘤胃和皱胃中相对表达最高,老年组和幼年组EGFR相对表达量较高,初生组相对表达量较低。上述结果表明,EGF和EGFR在牦牛各胃中的表达与年龄具有相关性,分布特点具有相似性,对牦牛各胃的发育及生理功能的实现具有重要调控作用,推测两者的特异性结合与促进胃黏膜上皮组织的增殖和修复以及抑制皱胃胃酸和胃蛋白酶的分泌等其他胃功能有关,并在牦牛各胃的生长发育过程中发挥功能。The aim of this experiment was to investigate the distribution and expression differences of epidermal growth factor(EGF)and epidermal growth factor receptor(EGFR)in the rumen,reticulum,omasum and abomasum of yaks of different ages,to investigate the relationship between EGF and EGFR and the structure and physiological function of the stomach of yaks.In this experiment,20 healthy yaks were selected and divided into newborn,juvenile,adult and senior groups,with 3 heads in each group,tissue and molecular samples were taken from the rumen,reticulum,omasum and abomasum at slaughter;The expression and distribution of EGF and EGFR were studied using quantitative real-time PCR,immunohistochemistry,and immunofluorescence,and the results were statistically analyzed.In result,the relative expression of EGF and EGFR was higher in the mucosal epithelium of the foregut(rumen,reticulum,omasum)of yaks in four age groups,high number of positive products in epithelial cells;Positive distribution in the parirtal cells of the fundic gland area and in the mucous neck cells in the abomasum,and the expression levels were significantly different in different age groups(P<0.05);Relative expression of EGF and EGFR was lowest in each stomach in senior agroup,highest in the rumen and abomasum in the newborn group,the relative expression of EGFR was higher in the senior and juvenile groups and lower in the newborn group.The above results indicated that the expression of EGF and EGFR in the stomachs of yaks is age-dependent and their distribution characteristics are similar,and they play an important role in regulating the development and physiological functions of the stomachs of yaks.It is hypothesized that their specific binding is related to promoting the proliferation and repair of gastric mucosal epithelial tissues and inhibiting the secretion of gastric acid and pepsin in the wrinkled stomach,among other gastric functions,and that they function in the growth and development of the stomachs of yaks.
