摘 要:目的 研究玉米黄素双棕榈酸酯经大鼠口服后在胃肠道内容物、粪便中的含量及存在形式,为功效评价提供依据。方法 24只SD大鼠分为空白组和口服玉米黄素双棕榈酸酯组(低剂量组0.1 mL/100 g、中剂量组0.2 mL/100 g、高剂量组0.4 mL/100 g),饲养90 d后分别收集大鼠粪便、胃内容物、空肠和回肠段内容物,采用高效液相色谱法(HPLC)测定玉米黄素和玉米黄素双棕榈酸酯。结果 大鼠口服玉米黄素双棕榈酸酯后,胃内容物中含有玉米黄素双棕榈酸酯(100~300μg·g^(-1)),与给药剂量呈正比;空肠中玉米黄素和玉米黄素双棕榈酸酯的含量是胃内容物中的2倍左右;回肠内容物中,高剂量组玉米黄素的含量是低、中剂量组的100倍;粪便中玉米黄素双棕榈酸酯的含量为玉米黄素的10倍左右。结论 玉米黄素双棕榈酸酯在胃内不易分解,受肠道内环境、肠道酶的影响在肠道内容物中部分转换为玉米黄素,剩余大量药物则以原形在粪便中被排出体外。Objective To investigate the content and form of zeaxanthin dipalmitate in the gastrointestinal tract and feces of SD rats after oral administration,and to provide a basis for the efficaoy evaluation.Methods A total of 24 rats were bred for 90 days and divided into blank group and oral zeaxanthin di-palmitate group(low-dose group 0.1 mL/100 g,medium-dose group 0.2 mL/100 g,high-dose group 0.4 mL/100 g).Feces,stomach contents,jejunum and ileum contents were determined by HPLC method for zeaxanthin and zeaxanthin dipalmitate.Results After oral administration of zeaxanthin dipalmitate by rats,the gastric content species containing zeaxanthin dipalmitate(100-300μg·g^(-1))was proportional to the dose;the content of zeaxan-thin dipalmitate and zeaxanthin in the jejunum was twice that of the gastric content.In the contents of the ileum,the content of zeaxanthin in the high-dose group was 100 times that of the low-dose and middle-dose groups;the content of zeaxanthin dipalmitate in feces was about 10 times that of zeaxanthin.Conclusion Zeaxanthin dipalmitate is not easily decomposed in the stomach,and may be affected by the intestinal environment and intestinal enzymes.Zeaxanthin dipalmitate is converted into a certain amount of zeaxanthin in the intestinal contents,and a large amount of zeaxanthin dipalmitate is excreted from the feces in its original form.
