摘 要:目的分析沉默细胞因子信号抑制因子1(SOCS1)对肝细胞癌患者树突状细胞表面抗原及其免疫调控的影响。方法采集宁夏回族自治区人民医院2020年10月至2021年10月收治的60例肝细胞癌患者的外周静脉血,通过淋巴细胞分离液离心得到外周血单核细胞,进行相关细胞表面标志物水平、SOCS1蛋白表达、树突状细胞表面抗原及树突状细胞上清液中细胞因子水平检测。结果流式细胞术检测CD3^(+)细胞约占94%,CD3^(+)CD4^(+)细胞约占36%,而CD3^(+)CD8^(+)细胞约占52%。CD1a和CD83分别为树突状细胞特异性和成熟的细胞标记物,CD1a^(+)细胞约占93%,CD1a^(+)CD83^(+)细胞约占85%。在诱导0、1、3 d时miR-618表达水平无明显变化,在第5、7天miR-618表达水平均降低(P均<0.05)。双荧光素酶系统表明,miR-618与SOCS1表达具有靶向调控作用(P<0.05)。miR-618过表达后,SOCS1蛋白表达量低于mimic NC组(P<0.05)。与各对照组相比,miR-618 mimic组和SOCS1 shRNA组CD11c、HLA-DR和CD86阳性细胞均增多(P均<0.05),CD4阳性细胞无明显变化(P>0.05)。miR-618 mimic^(+)sh-SOCS1组CD11c、HLA-DR、CD86阳性细胞数均增多(P均<0.05)。与各对照组相比,miR-618mimic组和shRNA-SOCS1组TNF-γ、IL-12和IFN-α表达水平均上升(P均<0.05),miR-618 mimic^(+)sh-SOCS1组上述因子均增加(P均<0.05)。结论沉默SOCS1可以促进肝细胞癌患者树突状细胞活化,提高其免疫调控能力。Objective To analyze the effect of silencing suppressor of cytokine signaling 1(SOCS1)on dendritic cell surface antigen and its immune regulation in patients with liver cancer.Methods The peripheral blood of 60 patients with liver cancer in People’s Hospital of Ningxia Hui Autonomous Region from October 2020 to October 2021 were collected.Peripheral blood mononuclear cells were obtained by lymphocyte separation solution centrifugation,and the levels of related cell surface markers,SOCS1 protein expression,dendritic cell surface antigen detection,and cytokine levels in dendritic cell supernatant were detected.Results Flow cytometry detected about 94%of CD3^(+)cells,36%of CD3^(+)CD4^(+)cells,while about 52%of CD3^(+)CD8^(+)cells.CD1a and CD83 were dendritic cell-specific and mature cell markers,respectively,with about 93%of CD1a^(+)cells and 85%of CD1a^(+)CD83^(+)cells.There was no significant change in miR-618 expression levels at induction 0,1 and 3 d.The miR-618 expression levels were significantly lower on days 5 and 7 compared to the previous days(P all<0.05).The dual luciferase system demonstrated a targeting regulatory relationship between miR-618 and SOCS1 expression(P<0.05).miR-618 overexpression resulted in significantly lower SOCS1 protein expression than mimic NC(P<0.05).Compared with each control group,CD11c,HLA-DR and CD86 positive cells were significantly increased in miR-618 mimic group and SOCS1 shRNA group(P all<0.05),but there was no significant change in CD4 positive cells(P>0.05).The numbers of CD11c,HLA-DR,CD86 positive cells were significantly increased in miR-618 mimic+sh-SOCS1 group(P all<0.05).The expression levels of TNF-γ,IL-12 and IFN-αwere significantly increased in the miR-618 overexpression group and SOCS1 silencing group compared with the control of each group(P all<0.05),and the increase of the above factors were more significant in the miR-618 mimic^(+)sh-SOCS1(P all<0.05).Conclusion Silencing SOCS1 could promote the activation of dendritic cells in patients with liver cancer
