摘 要:目的:探讨不同浓度臭氧水(OW)对人牙龈成纤维细胞(HGFs)的细胞毒性。方法:体外培养HGFs,按干预臭氧水浓度分为5组:1、2、4、6 mg/L和空白对照组,在作用的不同时点(1、5、10、20 min)采用倒置相差显微镜观察各组细胞的形态变化,MTT法测定细胞的相对增殖率,采用SPSS软件进行统计分析。结果:浓度为1和2 mg/L的OW在20 min内对HGFs的细胞毒性为1级,4 mg/L作用1 min,细胞毒性为1级,作用5~20 min,细胞毒性为2级;6 mg/L作用1~20 min,细胞毒性为2~3级。结论:臭氧水的细胞毒性与其浓度和作用时间有关,低浓度臭氧水(≤2 mg/L)可应用于临床治疗和消毒中,在保证作用效果的情况下,应尽量减小臭氧水的作用时间和浓度。Objective:To assess the cytotoxicity of various concentrations of ozonated water(OW)on human gingival fibroblas(HGFs).Methods:HGFs were cultured with enzyme digestion and trypsinization.The cells were divided into 5 groups and exposed to OW with concentration(mg/L)of 1,2,4,6 and 0(control)respectively.Cytotoxicity was evaluated after the exposure for 1,5,10 and 20 min respectively.Surface morphology and histologic changes were observed by the inverted phase contrast microscope.The cytotoxicity of OW was evaluated by MTT assay.SPSS software was adopted for statistical analysis.Results:The cytotoxicity of OW at 1 and 2 mg/L with an exposure of 20 min or less was grade 1,that at 2 with the exposure of 1 min was grade 1 and with 5-20 min was grade 2,that at 6 mg/L with 1-20 min exposure was grade 2-3.Conclusion:The cytotoxicity of ow increases with the concentration increase and the extended exposure time.OW of 2 mg/L might serve as a possible adjuvant in clinical treatment and disinfection.The exposure time should be reduced as far as possible.
