摘  要:目的 评价天冬的食用安全性,为申报食药物质提供科学依据。方法 在内江市选用SD大鼠、KM小鼠按限量法设10 000 mg/kg体重1个剂量组进行某天冬粉末急性经口毒性试验。在某天冬粉末的遗传毒性试验中,细菌回复突变试验设8、40、200、1 000和5 000μg/皿5个剂量组,另设未处理对照组、溶剂(蒸馏水)对照组、二甲基亚砜(DMSO)对照组和阳性对照组,试验在±S9条件下进行,每组作3个平行皿,重复1次;哺乳动物红细胞微核试验设1 000、2 000和4 000 mg/kg体重3个剂量组,另设阴性对照及阳性对照组;体外哺乳类细胞染色体畸变试验设1.25、2.50和5.00 mg/ml 3个剂量组,另设阴性对照组和阳性对照组,试验在±S9条件下进行。哺乳动物红细胞微核试验采用卡方检验,检验水准α为0.05;体外哺乳类细胞染色体畸变试验采用卡方检验,Bonferroni法修正后检验水平α为0.012 5。结果 天冬对SD大鼠、KM小鼠急性经口LD50值大于10 000 mg/kg体重;在±S9条件下,TA97a、TA100、TA98、TA102和TA1535这5种菌株在5个剂量组的平均回变菌落数均未超过溶剂(D-H2O)对照组的2倍,且无剂量-反应关系;3个剂量组雌性KM小鼠的含微核细胞率均值分别为3.0‰、2.2‰和2.9‰,与阴性对照组(均值3.0)相比较差异均无统计学意义(χ^(2)=0.000,P=1.000;χ^(2)=1.234,P=0.267;χ^(2)=0.017,P=0.896);3个剂量组雄性KM小鼠的含微核细胞率均值分别为1.5‰、1.5‰和1.7‰,与阴性对照组(均值2.1)相比较差异均无统计学意义(χ^(2)=1.002,P=0.317;χ^(2)=1.002,P=0.317;χ^(2)=0.422,P=0.516);在+S9条件下,3个剂量组的染色体总畸变率均为2%,与阴性对照组(1%)比较差异均无统计学意义(χ^(2)=0.338,P=0.561;χ^(2)=0.338,P=0.561;χ^(2)=0.338,P=0.561),在-S9条件下,3个剂量组的染色体总畸变率分别为1%、2%和3%,与阴性对照组(2%)比较差异均无统计学意义(χ^(2)=0.338,P=0.561;χ^(2)=0.000,P=1.000;χ^(2)=0.205,P=0.651)。结论 在本�Objective To evaluate the edible safety of Asparagus cochinchinensis,and to provide scientific basis for its declaration as food or drug substance.Methods The acute oral toxicity test of a certain Asparagus cochinchinensis powder from Neijiang City was performed on SD rats and KM mice with a dose group of 10000 mg/kg·BW.In the genotoxicity test of Asparagus cochinchinensis powder from Neijiang City,the bacterial reverting mutation test was performed in 5 dose groups(8,40,200,1000 and 5000μg/plate),and untreated control group,solvent(distilled water,D-H_(2)O)control group,dimethyl sulfoxide(DMSO)control group and positive control group were also set up.The test was conducted under±S_(9) condition with 3 parallel plates and repeated once.Mammalian erythrocyte micronucleus test was set up in 3 dose groups of 1000,2000 and 4000 mg/kg·BW,and negative control group and positive control group were also set up.In vitro mammalian cell chromosome aberration test was performed in 1.25 mg/ml,2.50 mg/ml and 5.00 mg/ml dose groups,respectively,as well as in negative control group and positive control group.The test was conducted under±S_(9) condition.The mammalian erythrocyte micronucleus test was performed by Chisquare test,and the test level wasα=0.05.The vitro mammalian cell chromosome aberration test was performed by Chi-square test,and the test level wasα=0.0125 after modified by Bonferroni method.Results The acute transoral LD_(50)(median lethal dose)value of Asparagus cochinchinensis in SD rats and KM mice was more than 10000 mg/kg·BW.Under the condition of±S_9,the average retrograde colony number of TA_(97a),TA_(100),TA_(98),TA_(102)and TA_(1535)strains in the five dose groups was not more than twice that of the solvent(D-H_2O)control group,and there was no dose-response relationship.The average micronucleus containing cell rates of female KM mice in the three dose groups were 3.0‰,2.2‰and 2.9‰,respectively,and there was no significant difference compared with the negative control group(mean 3.0)(χ^(2)=
