microRNA-4500靶向IGF2BP1抑制人胶质瘤细胞增殖、侵袭、迁移的机制研究

摘  要:目的探究microRNA-4500通过调控靶基因IGF2BP1对人胶质瘤细胞增殖、侵袭、迁移的影响。方法选取人胶质母细胞瘤细胞株U87MG和正常人脑胶质细胞系LN-18为实验细胞,采用RT-PCR法检测U87MG和LN-18中miR-4500、IGF2BP1的表达水平;分别采用miR-4500 NC、miR-4500 mimic、miR-4500 NC inhibitor、miR-4500 inhibitor转染U87MG和LN-18细胞;采用CCK8法检测U87MG细胞的增殖能力;采用Transwell实验检测U87MG细胞的侵袭和迁移能力;采用双荧光素酶报告基因检测miR-4500靶基因。结果与正常胶质细胞相比,胶质瘤细胞U87MG中miR-4500呈低表达(P<0.05),IGF2BP1则呈相对高表达(P<0.05)。与miR-4500 NC组细胞相比,miR-4500 mimic组胶质瘤细胞的增殖、迁移、侵袭能力明显增高(P<0.05),与miR-4500 NC inhibitor组细胞相比,miR-4500 inhibitor组胶质瘤细胞的增殖、迁移、侵袭能力则明显较低(P<0.05)。生物信息学网站预测及双荧光素酶报告基因实验结果显示,miR-4500与IGF2BP13′UTR存在结合位点。结论胶质瘤细胞中miR-4500颗通过抑制靶基因IGF2BP1表达促进胶质瘤细胞的增殖、迁移和侵袭。Objective To investigate the effect of microRNA⁃4500 on the proliferation,inva⁃sion and migration of human glioma cells by regulating the target gene IGF2BP1.Methods Human glioblastoma cell line U87MG and normal human brain glial cell line LN⁃18 were select⁃ed as experimental cells,and the expression levels of miR⁃4500 and IGF2BP1 in U87MG and LN⁃18 were detected by RT⁃PCR.MiR⁃4500 NC,miR⁃4500 mimic,miR⁃4500 NC inhibitor,and miR⁃4500 inhibitor were transfected into U87MG and LN⁃18 cells,respectively.The pro⁃liferation ability of U87MG cells was detected by CCK8 method.Invasion and migration of U87MG cells were detected by Transwell assay.Target genes of miR⁃4500 were detected by double luciferase reporter gene.Results Compared with normal glial cells,the expression of miR⁃4500 in U87MG glioma cells was low(P<0.05),while the expression of IGF2BP1 was relatively high(P<0.05).Compared with miR⁃4500 NC group,the proliferation,migration and invasion ability of glioma cells in miR⁃4500 mimic group was significantly increased(P<0.05),and compared with miR⁃4500 NC group,the proliferation,migration and invasion abil⁃ity of glioma cells in miR⁃4500 inhibitor group was significantly lower(P<0.05).Bioinforma⁃tics website prediction and double luciferase reporter gene results showed that there was a bind⁃ing site between miR⁃4500 and IGF2BP13′UTR.Conclusion MiR⁃4500 glioma cells promote the proliferation,migration and invasion of glioma cells by inhibiting the expression of target gene IGF2BP1.
