摘 要:目的:筛选缺血性卒中的差异表达基因及重要信号通路,为探索缺血性卒中的发生机制提供理论依据。方法:下载GEO数据库中有关缺血性卒中的基因测序,依照纳入标准筛选满足条件的数据集。用R软件对数据进行分析处理,获得有差异表达的基因;通过String数据库构建蛋白质相互作用(protein-protein interaction,PPI)网络;通过KOBAS数据库完成GO功能和KEGG富集分析。结果:筛选出缺血性卒中芯片数据GSE146882和GSE198710,二者有29个共同的差异表达基因,其中表达上调的基因有23个,表达下调的基因有6个。PPI网络构建结果显示,相互关联最紧密的6个基因分别为CXCL8、MYL6、RGS18、CPA3、AOAH和SUCLG2。富集分析结果提示,信号通路主要富集于代谢、蛋白质消化和吸收、氧化磷酸化等重要通路。结论:基于GEO数据库筛选出了与缺血性卒中可能相关的29个差异表达基因,并完成了PPI网络构建和功能富集分析,为探索缺血性卒中的机制提供了理论依据。Objective:To screen differentially expressed genes and important signaling pathways in ischemic stroke and provide a theoretical basis for exploring the pathogenesis of ischemic stroke.Methods:The gene sequencing of ischemic stroke in GEO database was downloaded,and the eligible data sets were chosen according to the inclusion criteria.Data were analyzed with R software to obtain differentially expressed genes.Protein-protein interaction(PPI)network was constructed through String database.GO function and KEGG enrichment analysis were completed through KOBAS database.Results:The ischemic stroke microarray data GSE146882 and GSE198710 were screened out,which had 29 common differentially expressed genes,including 23 up-regulated genes and 6 down-regulated genes.The results of PPI network showed that the 6 most closely related genes were CXCL8,MYL6,RGS18,CPA3,AOAH and SUCLG2.The results of enrichment analysis suggested that signaling pathways were mainly enriched in metabolism,protein digestion and absorption,oxidative phosphorylation and other important pathways.Conclusion:Based on GEO database,twenty-nine differentially expressed genes possibly associated with ischemic stroke are screened,PPI network construction and functional enrichment analysis are completed,providing a theoretical basis for exploring the mechanism of ischemic stroke.
