摘 要:高盐废水指含盐质量分数大于3%(以NaCl计)的废水,通常来自海水养殖业、食品工业、医药行业等。水体较高的盐度可对微生物的脱氮活性形成抑制,导致反硝化细菌效能降低。从青海省察尔汗盐湖沉积土样品中筛选到一株耐盐反硝化细菌,命名为NA2-1。通过形态特征观察、生理生化指标和16S rRNA基因序列分析,初步认定菌株NA2-1为盐渍微菌(Salimicrobium sp.)。进一步对菌株NA2-1生长条件进行优化,得到该菌株的最适生长条件为:温度35℃,盐度4%,pH为5.5,碳源为红糖,氮源为酵母粉。在盐度为4%、初始氮质量浓度为61.34 mg/L的反硝化培养基中,48 h内对硝态氮去除率达到91.3%。另外,菌株NA2-1在盐度为3%~15%的培养基内生长良好,具有良好的耐盐能力;在15%盐度下,该菌株具有较高的四氢嘧啶产量,可达到0.74 mg/mL。菌株NA2-1具有良好的除氮效果,对高盐环境具有良好的耐受性,可为生物除氮提供微生物资源。High salinity wastewater refers to wastewater containing more than 3%salt(in terms of NaCl),usually from mariculture,food industry,pharmaceutical industry,etc.High salinity can cause inhibition of the denitrification activity of microorganisms and lead to a decrease in the effectiveness of denitrifying bacteria.A strain of salt-tolerant denitrifying bacteria NA2-1 was screened from the sediment soil sample of Chalhan Salt Lake,Qinghai Province,and was initially identified as Salimicrobium sp.by morphological characteristics observation,physiological and biochemical indicators and 16S rRNA gene sequence analysis.The optimal growth conditions for strain NA2-1 were further optimized and obtained as follows:temperature 35℃,salinity 4%,pH 5.5,carbon source brown sugar and nitrogen source yeast powder.In the denitrification medium with a salinity of 4%and an initial nitrogen mass concentration of 61.34 mg/L,91.3%of nitrate-nitrogen was removed within 48 h.In addition,strain NA2-1 strain could grow well in the medium with salinity ranging from 3%to 15%and had good salt tolerance.At 15%salinity,the strain had high ectoine production,which could reach 0.74 mg/mL.Strain NA2-1 has good nitrogen removal effect and good tolerance to high salinity,which can provide a microbial resource for biological nitrogen removal.
