摘  要:目的 探究红景天苷是否能够通过调控PTEN/AKT/mTOR通路增强自噬,从而发挥对帕金森病(PD)模型小鼠的治疗作用。方法 24只C57BL/6小鼠随机分为空白组、模型组与红景天苷组,每组8只。转棒实验评估小鼠的运动协调性和平衡能力;免疫荧光染色法检测脑黑质区酪氨酸羟化酶(TH)和α-突触核蛋白(α-Syn)表达情况;试剂盒测定脑黑质区线粒体ATP酶(ATPase)、线粒体呼吸链复合物Ⅰ(Complex I)活性;Western blot测定人第10号染色体缺失的磷酸酶(PTEN)诱导假定激酶1(PINK1)、泛素连接酶parkin、微管相关蛋白1轻链3(LC3)、核孔糖蛋白62(p62)表达、PTEN、蛋白激酶B(Akt)、磷酸化-Akt(p-Akt)、哺乳动物雷帕霉素靶蛋白(mTOR)及p-mTOR蛋白表达水平。结果 与模型组相比,红景天苷组小鼠旋转圈数明显减少、掉落潜伏期明显延长;TH表达明显增加、α-Syn表达明显降低;ATPase、Complex I活性明显提高;PINK1和parkin表达及LC3-II/LC3-I比值明显升高,而p62表达明显降低;PTEN蛋白表达明显增加,而p-Akt、p-mTOR蛋白表达水平明显降低(P<0.05)。结论 红景天苷能够通过调控PTEN/AKT/mTOR通路活性,增强自噬水平,从而起到治疗PD模型小鼠的作用。Objective To explore whether salidroside can enhance autophagy by regulating the PTEN/AKT/mTOR pathway,and play a therapeutic role in Parkinson's disease(PD)model mice.Methods Twenty-four C57BL/6 mice were randomly divided into control group,model group and salidroside group,with 8 mice in each group.The motion coordination and balance ability of mice were evaluated by rotary rod test.The expressions of Tyrosine hydroxylase(TH)andα-synuclein(α-Syn)in the substantia nigra were observed by immunofluorescence staining.The activities of mitochondrial ATPase and mitochondrial respiratory chain Complex I(Complex I)in the substantia nigra were determined by the kit.The expressions of phosphatase and tensin homolog deleted on chromosome ten(PTEN)-induced putative kinase(PINK1),ubiquitin ligase(Parkin),microtubule-associated protein 1 light chain 3(LC3),nucleoprotein 62(p62),PTEN,protein kinase B(Akt),phosphorylated-Akt(p-Akt),mammalian target of rapamycin(mTOR)and p-mTOR were detected by Western blot.Results Compared with model group,the rotation number of mice in salidroside group was significantly reduced and the falling latency was significantly prolonged.The expression of TH was increased significantly.The expression ofα-Syn was significantly decreased.The activities of ATPase and Complex I were significantly increased.The expressions of PINK1 and Parkin and LC3-I/LC3-I ratio were significantly increased,while expression of p62 was significantly decreased.The expression of PTEN was significantly increased,and the expressions of p-Akt and p-TOR was significantly decreased(P<O.05).Conclusion Salidroside can enhance autophagy by regulating the activity of PTEN/AKT/mTOR pathway,thus playing a role in the treatment of PD model mice.
