摘  要:目的 分析EZH2对MDA-MB-231三阴性乳腺癌细胞生物学行为的调节能力。方法 通过质粒转染的方法过表达和沉默MDA-MB-231三阴性乳腺癌细胞中的EZH2;通过MTT分析EZH2对细胞增殖的影响;通过流式细胞检测分析EZH2对细胞凋亡和周期的影响;通过Transwell分析EZH2对细胞侵袭能力的影响。结果 EZH2的过表达能够促进MDA-MB-231三阴性乳腺癌细胞的增殖,增加处于分裂期细胞的比例,抑制细胞凋亡,促进细胞侵袭;EZH2的沉默能够抑制MDA-MB-231三阴性乳腺癌细胞的增殖,减少处于分裂期细胞的比例,促进细胞凋亡,抑制细胞侵袭。结论 EZH2具有调节MDA-MB-231三阴性乳腺癌细胞增殖、周期、凋亡和侵袭的能力。Objective To analyze the regulatory ability of EZH2 on the biological behavior of MDA-MB-231 triple-negative breast cancer cells.Methods Overexpress and silence EZH2 in MDA-MB-231 triple-negative breast cancer cells by plasmid transfection.The effect of EZH2 on cell proliferation was analyzed by MTT method.The effect of EZH2 on cell apoptosis and cycle was analyzed by flow cytometry.The impact of EZH2 on cell invasion was analyzed by Transwell.Results Overexpression of EZH2 can promote the proliferation of MDA-MB-231 triple-negative breast cancer cells,increase the proportion of cells in the dividing phase,inhibit cell apoptosis,and promote cell invasion.Silencing of EZH2 can inhibit the proliferation of MDA-MB-231 triple-negative breast cancer cells,reduce the proportion of cells in the dividing phase,promote cell apoptosis and inhibit cell invasion.Conclusion EZH2 has the ability to regulate the proliferation,cycle,apoptosis and invasion of MDA-MB-231 triple-negative breast cancer cells.
