摘 要:目的 探讨circ_0060927对结直肠癌细胞凋亡、迁移和侵袭的影响及作用机制。方法 收集31例结直肠癌患者的癌组织和对应的癌旁组织,RT-qPCR法检测组织中circ_0060927和miR-1225-5p表达水平。体外培养结直肠癌细胞SW620,分别转染si-circ_0060927、miR-1225-5p模拟物、共转染si-circ_0060927与anti-miR-1225-5p,分别采用流式细胞术、Transwell及Western blot检测沉默circ_0060927或过表达miR-1225-5p对SW620细胞凋亡、迁移和侵袭及细胞中Bax、Bcl-2、MMP-2和MMP-9蛋白表达的影响。双荧光素酶报告基因实验验证circ_0060927和miR-1225-5p调控关系。结果 与癌旁组织比较,结直肠癌组织中circ_0060927表达升高(P<0.05),而miR-1225-5p表达降低(P<0.05)。沉默circ_0060927或过表达miR-1225-5p后,SW620细胞凋亡率及细胞中Bax蛋白表达升高(P<0.05),迁移和侵袭数及细胞中Bcl-2、MMP-2和MMP-9蛋白表达降低(P<0.05)。与仅沉默circ_0060927的SW620细胞比较,同时沉默circ_0060927和miR-1225-5p的SW620细胞凋亡率及细胞中Bax蛋白表达降低(P<0.05),迁移和侵袭数及细胞中Bcl-2、MMP-2和MMP-9蛋白表达升高(P<0.05)。双荧光素酶报告基因实验结果显示,circ_0060927可靶向结合miR-1225-5p;同时沉默circ_0060927促进了SW620细胞中miR-1225-5p的表达。结论 circ_0060927在结直肠癌组织中表达升高,沉默其表达可能通过靶向上调miR-1225-5p促进结直肠癌细胞SW620凋亡,并抑制细胞迁移和侵袭。Objective To investigate the effect and mechanism of circ_0060927 on the apoptosis,migration and invasion of colorectal cancer cells.Methods The cancer tissues and corresponding adjacent tissues of 31 patients with colorectal cancer were collected,and the expression levels of circ_0060927 and miR-1225-5p in the tissues were detected by RT-qPCR.Colorectal cancer cell line SW620 was cultured in vitro.After si-circ_0060927 or miR-1225-5p mimics was transfected into SW620 cells,or si-circ_0060927 and anti-miR-1225-5p were co-transfected into SW620 cells,flow cytometry,Transwell and Western blot were used to detect the effects of silencing circ_0060927 or overexpressing miR-1225-5p on SW620 cell apoptosis,migration and invasion,and the protein expressions of Bax,Bcl-2,MMP-2 and MMP-9 in the cells,respectively.The dual luciferase reporter gene experiment verified the regulatory relationship between circ_0060927 and miR-1225-5p.Results Compared with the adjacent tissues,the expression of circ_0060927 in colorectal cancer tissues was increased(P<0.05),while the expression of miR-1225-5p was decreased(P<0.05).After silencing circ_0060927 or overexpressing miR-1225-5p,the apoptosis rate and the expression of Bax in SW620 cells were increased(P<0.05),while the number of migration and invasion,and the expressions of Bcl-2,MMP-2 and MMP-9 in cells were decreased(P<0.05).Compared with SW620 cells that only silenced circ_0060927,the apoptosis rate and the expression of Bax in SW620 cells that silenced circ_0060927 and miR-1225-5p at the same time decreased(P<0.05),while the number of migration and invasion,and the expressions of Bcl-2,MMP-2 and MMP-9 increased(P<0.05).The results of the dual luciferase reporter gene experiment showed that circ_0060927 could target miR-1225-5p,while silencing circ_0060927 promoted the expression of miR-1225-5p in SW620 cells.Conclusion The expression of circ_0060927 is elevated in colorectal cancer tissues.Silencing its expression may promote the apoptosis of colorectal cancer cells Sw620 and in
