摘　　要：目的 探究脱嘌呤脱嘧啶核酸内切酶1(APE1)是否通过依靠调节肿瘤免疫微环境影响胃癌细胞SCC-7901生长。方法 通过GEPIA数据库分析胃癌组织和正常组织中APE1表达量。体外培养胃癌SCC-7901细胞,并分为APE1-inhibitor组和NC-inhibitor组,RT-PCR、Western blot分别检测细胞中APE1 mRNA及蛋白表达确定敲低效果;CCK8、Transwell实验分别检测细胞增殖、转移能力。取C57BL/c小鼠及免疫缺陷NCG小鼠构建胃癌腹腔移植瘤模型,接种转染APE1-inhibitor或NC-inhibitor的SCC-7901细胞,接种2周后观察小鼠肿瘤生长情况。结果 胃癌组织中APE1表达升高;下调APE1表达对体外培养胃癌细胞的增殖、转移能力无明显影响;下调APE1表达能抑制普通C57BL/c胃癌小鼠体内肿瘤生长,而对免疫缺陷胃癌小鼠肿瘤生长无明显影响。结论 APE1能促进胃癌细胞生长,但此过程可能依赖于体内肿瘤免疫微环境。Objective To investigate whether apurinic apyrimidinic endonuclease 1(APE1)affects the growth of gastric cancer cell line SCC-7901 by regulating tumor immune microenvironment.Methods The expression of APE1 in gastric cancer tissues and normal tissues was analyzed by GEPIA database.Gastric cancer SCC-7901 cells were cultured in vitro and divided into the APE1-inhibitor group and the NC-inhibitor group.The expression of APE1 mRNA and protein in the cells were detected by RT-PCR and Western blot to determine the knockdown effect.CCK8 and Transwell assays were used to detect proliferation and metastasis of cells,respectively.C57BL/c mice and immunodeficient NCG mice were inoculated with SCC-7901 cells transfected with APE1-inhibitor or NC-inhibitor.The tumor growth of the mice was observed after 2 weeks of inoculation.Results APE1 expression was increased in gastric cancer.The down-regulated expression of APE1 had no significant effect on proliferation and metastasis of gastric cancer cells in vitro.Down-regulating the expression of APE1 can inhibit the tumor growth of ordinary C57BL/c gastric cancer mice,but had no significant effect on the tumor growth of immunodeficient gastric cancer mice.Conclusion APEl can promote the growth of gastric cancer cells,but this process may depend on the tumor immune microenvironment in vivo.
