摘 要:目的探讨胃癌患者癌组织EphA5表达与临床病理特征的关系及EphA5促进胃癌进展的机制。方法2019年8月—2022年3月海南医学院第一附属医院诊治胃癌患者50例,取手术切除癌组织及癌旁组织,采用实时荧光定量PCR法检测EphA5、血管内皮生长因子(VEGF)、白细胞介素-6(IL-6)mRNA相对表达量,根据癌组织EphA5 mRNA相对表达量中位数4.249将50例患者分为EphA5高表达组(≥4.249)25例和EphA5低表达组(<4.249)25例,分析癌组织EphA5表达与胃癌患者临床病理特征的关系;采用Pearson相关法分析胃癌组织EphA5 mRNA与VEGF、IL-6 mRNA相对表达量的相关性。对数生长期SGC-7901细胞分为oe-NC组(转染oe-NC)、oe-EphA5组(转染oe-EphA5)、sh-NC组(转染sh-NC)、sh-EphA5组(转染sh-EphA5)。取转染72 h时4组细胞,采用实时荧光定量PCR法检测EphA5、VEGF、IL-6 mRNA相对表达量,采用Western blot法检测细胞EphA5、VEGF、IL-6蛋白相对表达量,采用CCK-8实验检测培养48 h时细胞增殖率,采用Transwell小室实验检测细胞侵袭数目。结果胃癌组织EphA5、VEGF、IL-6 mRNA相对表达量(4.74±2.54、3.27±1.25、5.23±2.74)均高于癌旁组织(1.00±0.62、1.00±0.46、1.00±0.54)(P<0.05)。胃癌组织EphA5 mRNA与VEGF、IL-6 mRNA相对表达量均呈正相关(r=0.395,P<0.001;r=0.453,P<0.001)。EphA5高表达组临床分期Ⅲ~Ⅳ期、有淋巴结转移比率(80.0%、68.0%)均高于EphA5低表达组(32.0%、28.0%)(χ^(2)=11.453,P=0.011;χ^(2)=0.366,P=0.024)。oe-EphA5组EphA5、VEGF、IL-6 mRNA及蛋白相对表达量均高于sh-EphA5组、oe-NC组、sh-NC组(P<0.05),sh-EphA5组均低于sh-NC组、oe-NC组(P<0.05);oe-EphA5组培养48 h时细胞增殖率(268.75±36.85)高于sh-EphA5组(175.86±37.74)、oe-NC组(224.56±41.86)、sh-NC组(229.75±32.58)(P<0.05),sh-EphA5组低于sh-NC组、oe-NC组(P<0.05);oe-EphA5组细胞侵袭数目[(174.75±18.57)个]多于sh-EphA5组[(42.58±9.35)个]、oe-NC组[(98.38±12.75)个]、sh-NC组[(102.57±16.84)个](P<0.05),sh-EphA5�Objective To investigates the relationships of the expression of EphA5 in cancer tissues with clinicopathological features and prognosis of patients with gastric cancer,and to explore the mechanism of EphA5 in the progression of gastric cancer.Methods Fifty patients with gastric cancer underwent surgical resection of cancer tissues and adjacent tissues in the First Affiliated Hospital of Hainan Medical University from August 2019 to March 2022.The relative expressions of EphA5,vascular endothelial growth factor(VEGF)and interleukin-6(IL-6)mRNAs were detected by real-time fluorescence quantitative PCR.According to the median relative expression level of EphA5 mRNA in cancer tissues(4.249);50 patients were equally divided into EphA5 high-expression group(≥4.249)and EphA5 low-expression group(<4.249).The relationship between EphA5 expression in cancer tissues and clinicopathological features of patients with gastric cancer was analyzed.The correlations of EphA5 mRNA in cancer tissues with the relative expressions of VEGF and IL-6 mRNA were evaluated by Pearson's correlation.The SGC-7901 cells in logarithmic growth phase were divided into oe-NC,oe-EphA5,sh-NC and sh-EphA5 groups,which were transfected with oe-NC,oe-EphA5,sh-NC and sh-EphA5,respectively.After 72-h transfection,the relative expressions of EphA5,VEGF and IL-6 mRNAs and proteins were detected by real-time fluorescence quantitative PCR and Western blot.The cell proliferation was assessed at 48 h by CCK-8 assay and the number of invasive cells was measured by Transwell assay.Results The relative expressions of EphA5,VEGF and IL-6 mRNAs were higher in cancer tissues(4.74±2.54,3.27±1.25,5.23±2.74)than those in adjacent tissues(1.00±0.62,1.00±0.46,1.00±0.54)(P<0.05).EphA5 mRNA in cancer tissues was positively correlated with the relative expressions of VEGF and IL-6 mRNAs(r=0.395,P<0.001;r=0.453,P<0.001).The percentages of patients with clinical stageⅢ-Ⅳand lymph node metastasis were higher in EphA5 high-expression group(80.0%,68.0%)than those in
